人疱疹病毒6型感染与口腔鳞癌相关性的研究

摘　　要：目的 :研究人疱疹病毒 6型 (HHV 6)感染与口腔鳞癌的关系。方法 :用间接免疫荧光试验及聚合酶链反应 ,分别检测口腔鳞癌和对照组患者血清中抗HHV 6IgG及外周血单个核细胞中HHV 6DNA序列 ;用免疫组化染色法 ,检测口腔鳞癌组织标本中HHV 6抗原。结果 :16例口腔鳞癌患者和 16例其他口腔疾病患者血清中抗HHV 6IgG的阳性率分别为 10 0 .0 %和 75 .0 % ,几何平均滴度分别为 1∶118和 1∶64 ,差异有显著意义 (P <0 .0 5 )。上述 2组患者HHV 6DNA的检出率分别为 62 .5 %和 12 .5 % (P <0 .0 5 )。其中 12例口腔鳞癌组织标本经免疫组化染色检测 ,9例 ( 75 % )HHV 6抗原阳性 ,而 8例对应癌旁组织中 ,阳性者仅 2例( 2 5 % ) ,两者间差异有显著性 (P <0 .0 5 )。结论 :HHV 6可能是口腔鳞癌发生的诱因 。Objective: To study the relationship between human herpes vi rus 6 (HHV-6) infection and oral squamors cell carcinoma. Methods: The serum anti-HHV-6 antibody titers from 16 cases of oral squamors cell carcinoma (OSCC) and 16 control subjects were detected by indirect immunofluores cence assay. HHV-6 DNA in peripheral blood mononuclear cells was amplified by P CR and the specificity was confirmed by Southern-blot hybridization with an int ernal probe oligonuclotide. An immunohistochemical staining was used to detect H HV-6 antigen in OSCC tissues. Results: Positive expression of s erum HHV-6 IgG antibody was found in all the cases of OSCC and in 12 of the 16 controls. Geometric mean titer of OSCC group and control group was 1∶118 and 1 ∶64 respectively (P<0.05), HHV-6 DNA in peripheral blood mononuclear cells were detected in 10/16 of OSCC cases and 2/16 of the controls respectively (P <0.05). 9 out of 12 OSCC cases and 2 out of 8 pericancerous cases were posit ive for HHV-6 antigens (P<0.05). Conclusion:HHV-6 possibly play a role in the pathogenesis of oral squamors cell carcinoma.
