摘  要:目的 :探讨不同剂量的三苯氧胺 (TAM)对实验性乳腺增生和血管形成的影响。方法 :用雌二醇和黄体酮诱导Wistar大鼠乳腺增生动物模型。 180只雌性大鼠随机分为 6组 :A组 :空白对照组 ,B组 :模型对照 (单用性激素 )组 ,C组 :性激素加 5倍TAM组 ,D组 :性激素加 10倍TAM组 ,E组 :性激素加15倍TAM组 ,F组 :性激素加 2 0倍TAM组 ,喂养 2个月后 ,检测DNA、AgNORS、Ⅷ因子及PCNA。结果 :1)各个TAM剂量组对细胞核DNA合成均有抑制作用 ,10倍剂量组已很明显 ;2 ) 10倍及以上TAM剂量组有明显抑制AgNORS计数的作用 ;3) 10倍及以上TAM剂量组有明显降低MVD的作用 ;4 ) 2 0倍TAM剂量组对PCNA表达有抑制作用。结论Objective To study the effect of different dosage of tamoxifen (TAM) on the experimental mammary hyperplasia and angiogenesis.Methods The animal model of mammary hyperplasia of Wistar rats was induced with estradiol and progesterone.One hundred and eighty female Wistar rats were selected randomly into Group A (control group ),Group B (model control group),Group C (5 fold dosage of TAM),Group D (10 fold dosage of TAM),Group E (15 fold dosage of TAM),and Group F (20 fold dosage of TAM)for 2 months.After that,samples of breasts were collected for the detection of DNA,AgNORS,Factor Ⅷ and proliferting nuclear cell antigen (PCNA).Results 1) All dosage groups of TAM had the inhibitory effect on synthesis of DNA,especially 10 fold dosage of TAM was the best of all.2) TAM group with more than 10 fold dosage had the obvious inhibitory effect on the AgNORS count.3) TAM group with more than 10 fold dosage had the effect of obviously decreasing microvessel density (MVD).4) Only 20 fold dosage of TAM had the inhibitory effect on the expression of PCNA.Conclusion TAM(especially 10 fold dosage) is proved to have the obvious inhibitory effect on mammary hyperplasia and angiogenesis.
