结肠癌nm23-H1和CD_(44)V_6及PCNA表达与DNA含量及转移潜能的相关性

摘　　要：目的 :探讨nm2 3 H1、CD44V6和PCNA和DNA含量在结肠癌生长、浸润、转移中的作用及其相互关系。方法 :采用免疫组化S P方法检测 15 2例结肠癌nm2 3 H1、CD44V6和PCNA。同时采用流式细胞光度术测定 6 0例结肠癌DNA含量。结果 :15 2例结肠癌nm2 3 H1阳性率与淋巴结转移 ,P <0 0 5、癌组织分化关系密切 ,P <0 0 1;CD44V6表达与Dukes分期 (P <0 0 5 )、肿瘤浸润深度 (P <0 0 5 )、癌组织分化 (P <0 0 5 )及淋巴结转移 (P <0 0 1)密切相关 ;PCNA表达与Dukes分期及肿瘤浸润深度有关 (P<0 0 5 ) ;nm2 3 H1和CD44V6表达 ,两者呈负相关 (P <0 0 1) ,CD44V6阳性nm2 3 H1阴性表达的患者淋巴结转移率高于CD44V6阴性nm2 3 H1阳性表达者 (P <0 0 5 )。伴淋巴结转移的结肠癌CD44V6和PC NA共同阳性表达率和异倍体癌的发生率高于无转移组 (P <0 0 5 )。异倍体癌CD44V6和PCNA过度表达 ,而nm2 3 H1低表达 (P <0 0 5 )。结论 :nm2 3 H1、CD44V6和PCNA及DNA含量在结肠癌中的表达与其发生发展、侵袭转移密切相关 ,尤其CD44V6和nm2 3 H1表达在结肠癌转移中起协调作用 。Objective To explore the effect of nm23-H1,CD 44V 6,PCNA and DNA content in the metastasis of colonic carcinoma and their relation each other.Methods The expression of nm23-H1,CD 44V 6,PCNA in 152 cases of colonic carcinomas were studied immunohistochemically.DNA content in 60 cases were detected by Flow Cytometry.Results Expression of nm23-H1 was significantly correlated with histologic differentiation (P<0.01),and inversely associated with lymph node memastasis in colonic carcinomas(P<0.05).Expression of CD 44V 6 was closely related with Dukes stage(P<0.05),invasive extent (P<0.05),histological differentiation(P<0.05)and lymph node metastasis(P<0.01).Expression of PCNA was related with Dukes stage and invasive extent(P<0.05).Expression of nm23-H1 was inversely associated with that of CD 44V 6(P<0.01).Patients with positive CD 44V 6 and negative nm23-H1 expression had higher rates of lymph node metastasis than that with negative CD 44V 6 and positive nm23-H1 expression(P<0.05).The percentage of DNA aneuploidy colonic carainoma in the groups of lymph node metastasis was higher than in those of no metastasis (P<0.05) Over expression of CD 44V 6,PCNA and low expression of nm23-H1 were in the DNA aneuploidytumor(P<0.05).Conclusions The expression of nm23-H1,CD 44V 6,PCNA and DNA concent correlated with the development,invasion and metastasis of colonic carcinoma.Specially expressions of nm23-H1 and CD 44V 6 plays a coordinated role in the lymph node metastasis and served as an important indicator of metastasis of colonic carcinoma.
