摘  要:目的探讨纤维化及正常肝组织中血管紧张素Ⅱ1型受体(AT1R)mRNA的表达情况。方法应用半定量逆转录聚合酶链反应法检测18例肝癌周围纤维化肝组织和12例正常肝脏组织中AT1RmRNA的表达,同时逆转录AT1R特异性片段和内参片段(GAPDH),以二者的积分吸光度比值作为该受体mRNA的相对表达量。结果纤维化肝组织AT1RmRNA的相对表达量明显高于正常肝组织(1.27±0.45vs0.71±0.21,P<0.01)。结论AT1R可能在肝纤维化的发生发展中起一定的作用。Objective To investigate the expressi on profile of angiotensin Ⅱtype 1 receptor (AT1R) mRNA in fibrotic and normal li ver tissues. Methods Semi-quantitative RT-PCR was used to detect the expression of AT1R mRNA in 18 specimens of fibrotic liver tissues adjacent to liver cancer s and 12 normal liver tissue specimens. Specific AT1R product and GAPDH product used as internal control were both amplified by RT-PCR, and the ratio of their integrated optical densities calculated to estimate the relative quantity of AT 1R mRNA expression. Results The relative mRNA expression of AT1R in fibrotic l iver tissues was significantly higher than that of normal liver tissues (1.27±0 .45 vs0.71±0.21, P<0.01). Conclusion AT1R may play an important role in the de velopment of hepatic fibrosis.
