摘　　要：目的分段克隆SARS冠状病毒PUMC2株的全基因组cDNA。方法以SARS冠状病毒PUMC2株基因组RNA为模板,用RT-PCR扩增cDNA片段,PCR产物经纯化后,连接入pGEM-T载体中,进行序列测定。结果获得了SARS冠状病毒PUMC2株基因组全长cDNA的分段克隆。结论SARS冠状病毒PUMC2株基因组全长cDNA分段克隆的获得,为SARS冠状病毒基因功能的研究和全长有感染性cDNA的克隆奠定了基础。To get the cDNA clones which cover the whole genome of SARS-CoV PUMC 2 strain.Methods Using the SARS-CoV PUMC 2 strain genomic RNA as the template,the cDNA fragments were amplified by RT-PCR,the PCR products were further purified and ligated into the pGEM-T vector,and all the clones obtained were sequenced.Results The cDNA clones which cover the whole genome of SARS-CoV PUMC 2 strain were obtained.Conclusions These cDNAs can be provided for the function study of SARS-CoV proteins and the construction of full-length infectious cDNA clo ne of SARS-CoV.
