摘 要:目的:测定大肠癌细胞的胸苷磷酸化酶(dThdPase)蛋白含量,探讨dThdPase与5’-脱氧氟尿苷(5’-DFUR)、5-氟尿嘧啶(5-FU)对大肠癌细胞的生长抑制作用的相互关系。方法:采用ELISA法对6株大肠癌细胞系LS174T、CloneA、Col0320、CX-1、Lovo和MIP101进行dThdPase蛋白定量分析,然后进行5’-DFUR、5-FU对6株大肠癌细胞的生长抑制实验。把大肠癌细胞分别接种于96孔培养板的每孔中培养24 h,分别加入2倍梯度稀释的5’-DFUR和5-FU.继续培养96 h后应用MTT分析分别测定5’-DFUR和5-FU对6株大肠癌细胞的半数有效剂量(IC50)。结果:除LS174T检出0.5 u/mg、Lovo检出8.9 u/mg的dThdPase蛋白外,其它4株癌细胞未检出。6株大肠癌细胞均对5-FU高度敏感,其半数有效浓度(IC50)分别为1.52~13.68μmol/L,而5’-DFUR对6株大肠癌细胞的IC50则分别为68.71~528.15μmoL/L,是5-FU的15倍-94倍,差异有显著性(P<0.01)。结论:6株大肠癌细胞很少有dThadPase蛋白表达;每一株大肠癌细胞对5’-DFUR的敏感性均明显低于5-FU。Objective: To detect the thymidine phosphorytase ( dThdPase ) level in 6 colorectal carcinoma cell lines, and evaluate the anticancer effect of 5'-deoxy-5-Fluorouridine (5'-DFUR) , and 5-fluorouracil (5-FU) on those cells. Methods; dThdPase level was measured quantitatively by ELISA on 6 colorectal carcinoma cell lines LS174T, Clone A, Colo320, CX-1, Lovo, and MIPI01. Then the drug sensitivity of each colorectal carcinoma cell to 5'-DFUR and 5-FU was estimated. Firstly, 2 × 103 cells were seeded into each well of 96-well plate and incubated at 37 ℃ for 24 hours, then 5'-DFUR or 5-FU was added at a 2-fold-serial dilution (77 μmol/L to 0. 3/μmol/L in 5-FU and 1600 μmol/L to 3. 1 μmol/L in 5'-DFUR. After incubation for 96 hours, MTT assay was performed, and the 50% Inhibition Concentration ( IC50 ) for each drug was calculated with the linear regression of logarithmic transformation. Results; 0. 5 unit/mg and 8. 9 unit/mg of dThdPase level were detected in LS174T and Lovo respectively, but no dThdPase protein was detected in other 4 colorectal carcinoma cell lines. The IC50 of 5-FU on 6 colorectal carcinoma cell ranged from 1.52 ~ 13.68 μmol/L, while that of 5'-DFUR were 68.71 ~528.15 μmol/ L, 15 to 94 times higher than those of 5-FU (P <0. 0001). Conclusion; There was little dThdPase expression in 6 colorectal carcinoma cell lines tested. The IC50 of 5'-DFUR in each of colorectal carcinoma cell was higher than 5-FU in same cells significantly (P< 0.0001 ).
