摘　　要：目的　研究硫代磷酸化修饰的血管内皮生长因子 (VEGF)反义寡核苷酸 (ASODN)对慢性红白血病细胞K 5 62VEGF表达的影响。方法　将终浓度分别为 10、2 0、3 0 μmol·L-1的VEGFASODN和错义序列与K 5 62细胞分别孵育 2 4、48h ,另设空白对照组。采用RT PCR检测VEGFmRNA的表达 ,免疫组织化学方法检测VEGF蛋白的表达。结果　VEGFA SODN对K 5 62细胞VEGF表达有明显的抑制作用 ,与错义序列组和空白对照组比较有显著性差异 (P <0 .0 5 )。错义序列组与空白对照组比较VEGF的表达无显著性差异 (P >0 .0 5 )。结论　VEGFASODN可下调慢性红白血病细胞K 5Objective To study the effect of VEGF fully phosporothioated antisense oligodeoxynucleotide(VEGF ASODN) on VEGF expressed in K?562 cells in vitro. Methods K?562 cells were incubated with VEGF ASODN(eventual concentration as follows:10、20、30?μmol·L -1) or scrambled sequence,compared with negative control. The expression of VEGF mRNA was measured by semi-quantitative RT-PCR. The expression of VEGF protein was measured by immunohistochemistry. Results The result showed that VEGF ASODN had obvious inhibitive effect on expression of VEGF mRNA in K?562 cells, compared with scrambled sequence and negative control (P< 0.05). VEGF mRNA of K?562 cell was inhibited after treated by VEGF ASODN, and the inhibitive effect was the most obvious after 24 hours. The effect was dose-dependent(P<0.05). Scrambled sequence groups had no significant difference compared with negative control groups(P>0.05). VEGF ASODN had obvious inhibitive effect on expression of VEGF protein, compared with scrambled sequence and negative control(P<0.05). Conclusion The expression of VEGF mRNA and protein in K?562 cells are down-regulated after treated by VEGF ASODN.
