血管生成抑制剂TNP-470抑制ACHN肾癌生长的实验研究

摘　　要：目的　用ACHN肾癌细胞株建立肾癌动物模型 ,观察血管生成抑制剂TNP 4 70的抗肿瘤生长和转移效应。方法　将ACHN肾癌细胞株 2 0× 10 6/ 0 2ml接种于BALB/c裸鼠背部皮下 ,将 16只裸鼠随机分为对照组和用药组 ,自第 3天起分别给予溶剂 (3%无水乙醇 +97%生理盐水 ) 0 2ml和TNP 4 70 (4 0mg/kg) ,隔日给药。第 31天断颈处死小鼠 ,测定两组皮下瘤重、体积、微血管密度 (microvesseldensity ,MVD)及肺转移率。 结果　两组皮下瘤重分别为 (6 30 15± 12 3 6 5 )mg和 (2 70 0 0± 6 2 35 )mg (P <0 0 1) ,皮下瘤体积分别为 (316 16± 6 0 2 8)mm3 和(12 6 5 0± 4 5 12 )mm3 (P <0 0 1)。MVD计数 (4 0 0倍视野 )分别为 12 0 0± 3 78和 5 6 5± 2 31(P <0 0 1)。两组肺转移率分别为 6 2 5 %和 0 % (P <0 0 5 )。结论　血管生成抑制剂TNP 4 70通过抑制肿瘤血管生成而发挥抗瘤效应。Objective To study the inhibitory effect of TNP-470 on tumor growth and metastasis in ACHN renal cell carcinoma. Methods ACHN renal cell carcinoma cells (2.0×106/0.2 ml) were inoculated subcutaneously to the backs of 16 BALB/c nude mice. The mice were randomized into 2 groups. From the 3rd day, the control group was administered with vehicle only (3 % ethanol+97 % normal saline) and the treated group were subjected to the administration of 40 mg/kg of TNP-470 S.C. qod. On the 31st day, the weight and volume of subcutaneous tumors were measured, the microvessel density of subcutaneous tumors was counted, and the foci of lung metastasis were detected. Results The tumor weight in the control group and the treated group was (630.15±123.65) mg and (270.00±62.35) mg, respectively (P<0.01). The tumor volume in the control group and the treated group was (316.16±60.28) mm3 and (126.50±45.12) mm3, respectively (P<0.01). The MVD in the control group and treated group was (12.00±3.78) and (5.65±2.31), respectively (P<0.01). The lung metastasis rate in the two groups was 62.5 % (5/8) and 0 (0/8), respectively (P<0.05). Conclusion The anti-tumor effect of TNP-470 was mediated by anti-angiogenesis function.
