胃癌组织中黑色素瘤抗原基因-1,3基因的表达及其意义

摘　　要：目的　了解胃癌组织中黑色素瘤抗原基因 (MAGE) 1、3基因的表达 ,并探讨其作为免疫治疗的靶抗原在胃癌中的应用价值。方法　采用RT PCR法检测 36例胃癌患者的肿瘤组织和相应的癌旁“正常”黏膜中MAGE 1、3基因的表达情况 ,并以 37例慢性胃炎患者的胃镜活检组织和 4株胃癌细胞株作为对照。结果　 36例胃癌组织中 ,MAGE 1表达阳性率为 72 .2 2 % (2 6 / 36例 ) ,MAGE 3表达阳性率为 6 9.4 4 % (2 5 / 36例 ) ,MAGE 1、3表达双阳性率为 5 2 .78% (19/ 36例 ) ,MAGE 1、3任一基因表达阳性者 32例 ,阳性率为 88.89% (32 / 36例 )。癌旁“正常”黏膜中MAGE 1、3基因表达阳性率分别为 4 7.2 2 % (17/ 36例 )和 4 4 .4 4 % (16 / 36例 ) ,MAGE 1、3表达双阳性率为 30 .5 6 % (11/ 36例 )。 37例慢性胃炎组织中 ,MAGE 1、3表达阳性率分别为 2 9.73% (11/ 37例 )和2 7.0 3% (10 / 37例 ) ,MAGE 1、3表达双阳性率为 8.11% (3/ 37例 )。胃癌组织MAGE 1、3基因表达阳性率均高于癌旁“正常”黏膜组织和慢性胃炎患者。 4株胃癌细胞株MAGE 1、3基因表达均为阳性。结论　基于MAGE 1、3基因在胃癌中的高表达率 ,可利用其作为靶分子进行免疫治疗 。Objective To evaluate the immunotherapeutic application of tumor-associated antigens melanoma antigen gene (MAGE)-1, and -3 as the targets for gastric cancer immunotherapy. We detected MAGE-1, and -3 genes expression in gastric cancer. Methods MAGE-1 and MAGE-3 genes expression were detected by RT-PCR in tumor tissues and its parallelism mucosa. As comparative study of MAGE-1,and -3 genes expression was carried out in 37 biopsied tissues from chronic gastritis and four gastric cancer cell lines. Results There were 72.22%(26/36) of 36 gastric cancer patients with MAGE-1 gene expression in tumor tissue, twenty five cases(69.44%) with MAGE-3 gene expression positive and 19 cases with both positive MAGE-1 and MAGE-3. There were 32(88.89%) cases with either MAGE-1 or MAGE-3 expression positive in tumor tissue. MAGE-1 and MAGE-3 gene expression positive rates were 47.22%(17/36), and 44.44(16/36) respectively in the “normal” mucosa of the gastric cancer patients. While, in the biopsy tissues from chronic gastritis, the MAGE-1 and MAGE-3 genes expression positive rates were 29.73%(11/37) and 27.03%(10/37) respectively. There were 18 (48.68%) patients with chronic gastritis with either MAGE-1 or MAGE-3 expression positive. MAGE-1 and MAGE-3 genes expression positive rates in tumor tissues were significantly higher than those in its parallelism mucosae and biopsied tissues from chronic gastritis. Conclusion MAGE-1 and MAGE-3 are highly expressed in gastric cancer, and maybe the targets for immunotherapy. These two genes may play useful parameters for the follow up of the chronic gastritis patients.
