摘  要:目的 :研究6 %羟乙基淀粉溶液对大鼠全脑缺血 -再灌注损伤的影响。方法 :84只雄性Wistar大鼠随机分为假手术组 (S组 ,n=20)、缺血 -再灌注组 (I组 ,n=32)和6 %羟乙基淀粉溶液组 (H组 ,n=32) ,其中I组和H组又各分为再灌注1,3 ,6 ,12h亚组。采用原位杂交组织化学技术检测脑血管内皮细胞ICAM_1mRNA的表达。结果:I组和H组再灌注1 ,3 ,6,12h组ICAM -1mRNA的表达均较S组对应时间点组高(P<0.01) ,H组再灌注各个时间点组ICAM -1mRNA的表达均较I组对应时间点组低(P<0.05)。结论 :在不改变血细胞比容的条件下 ,再灌注早期应用6 %羟乙基淀粉溶液治疗可降低脑血管内皮细胞表面ICAM -1mRNA表达 ,减轻脑缺血 -再灌注损伤。Objective:To investigate the effect of6%hydroxyethyl starch on global cerebral ischemia-reperfusion injury in rats.Methods:Eighty-four male Wistar rats weighting230~280g were randomly divided into3groups which included sham operation group(group S,n=20)and ischemia-reperfusion group(group I,n=32),and infusion with6%hydroxyethyl starch(HES)solution group(group H,n=32).Groups I and H were further divided into4subgroups which included1h,3h,6h and12h after the beginning of reperfusion.Global cerebral ischemia was produced by permanent occlusion of bilateral vertebral arteries and cross-clamping of bilateral common carotid arteries for10min and then clamping was released for reperfusion.In group H,infusion was performed at10min after beginning of reperfusion.HES(5μL/g)was infused into the left femoral vein within5min.Hematocrit was checked before and after infusion.The animals were sacrificed at designed time and brain tissue was removed.In situ hybridization histochemistry technique was used to evaluate ICAM-1mRNA expression.Results:Compared with group S,ICAM-1mRNA expression in groups I and H at1h,3h,6h and12h after reperfusion increased significantly(P<0.01).ICAM-1mRNA expression in group H at1h,3h,6h and12h after reperfusion was lower than that in group I(P<0.05).Conclusion:6%HES could reduce the increase in ICAM-1mRNA expression and brain injury induced by cerebral ischemia-reperfusion in rats significantly.
