摘  要:目的 了解α1肾上腺素受体 (α1 adrenergicreceptor,α1 AR)三种亚型α1A AR ,α1B AR和α1D ARmRNA在大鼠肺组织的表达位置及缺氧性肺动脉高压发病过程中大鼠肺组织α1 AR亚型mRNA相对表达量的改变情况。方法 采用雄性Wistar大鼠 5 5只 ,随机分为正常对照组 (n =10 ) ,缺氧 2周组 (n =13)、缺氧 4周组 (n =10 ) ,生理盐水组 (n =10 ) ,酚妥拉明组 (n =12 ) ,应用半定量逆转录聚合酶链反应 (RT PCR)法 ,检测各组大鼠肺组织α1 AR三种亚型mRNA的相对表达量。应用组织原位杂交技术检测α1 AR三种亚型在正常大鼠肺组织中的表达部位。结果  ( 1)随缺氧时间的延长 ,肺动脉压力升高。 ( 2 )各实验组大鼠肺组织中α1 AR亚型mRNA的表达均以α1A AR为主 ,α1B AR次之 ,α1D AR含量最少。 ( 3)随缺氧时间的延长 ,α1A AR和α1B AR两种亚型mRNA相对表达量升高 ,缺氧 2、4周组与对照组差异有显著性 ,缺氧 2、4周组间差异亦有显著性。 ( 4 )随缺氧时间的延长 ,α1D ARmRNA的相对表达量增多 ,但缺氧组与对照组相比差异无显著性。 ( 5 )酚妥拉明组与生理盐水组比较 ,α1A AR ,α1B AR和α1D AR三种亚型mRNA的相对表达量差异均无显著性。 ( 6 )正常大鼠外周肺组织中三种α1 AR亚型的mRNA主要表达于肺小动脉和小静脉?Objective During the development of hypoxic pulmonary hypertension, the quantity of the protein and mRNA of α 1 adrenergic receptor (α 1 adrenergic receptor,α 1 AR) in the lung tissue increased, while no particular reports were found about the change of the α 1 AR subtypes during that course. The study aimed to understand the quantity and location of α 1 AR subtypes mRNA expression in control and hypoxia rats,and the effect of phentolamine in hypoxic pulmonary hypertension. Methods Fifty five male Wistar rats were divided randomly into 5 groups: control group ( n =10),hypoxia 2 weeks( n =13), hypoxia 4 weeks( n =10),saline control group( n =10)and hypoxia 4 weeks plus phentolamine group( n =12).Semi quantitative reverse transcription and polymerase chain reaction (RT PCR) were used to examine the mRNA expression of α 1 AR subtypes in each group. In situ hybridization was also used to detect the location of the α 1 AR in lungs of normal rats. Results (1) The pulmonary artery pressure increased with the extension of hypoxia. (2) The results of RT PCR showed that the mRNA expression of α 1A AR was the most,α 1B AR the second and α ZD AR was the least in all of the groups. (3) The expression of α 1A AR and α 1B AR mRNA increased with the extension of hypoxia, and the expressions among groups showed difference. (4) The expression of α 1D AR also increased with the extension of hypoxia but no difference was found among groups. (5) No difference was found in mRNA quantity of all three subtypes between phentolamine group and hypoxia saline group. (6) In situ hybridization showed that mRNA of the three α 1 AR subtypes located mainly in the artery and venous smooth muscle cells and endothelial cells. Conclusions This study suggested that α 1 AR subtypes worked in the development of hypoxia pulmonary hypertension. More research on α 1 AR subtypes may help the clinical treatment of pulmonary hypertension.
