柴地合方对慢性应激抑郁模型大鼠免疫功能的影响

摘　　要：目的 :观察柴地合方对慢性应激抑郁模型大鼠多种免疫功能的影响 ,探讨其抗抑郁作用可能的部分机理。方法 :以多种复合刺激交替进行的方法建立慢性应激抑郁模型 ,并取脾脏进行淋巴细胞体外培养。称量脾脏和胸腺重量测定胸腺指数和脾指数 ,以3 H胸腺嘧啶 (3 H TdR)掺入法检测T、B淋巴细胞增殖 ,以放射免疫法测定体外培养淋巴细胞的白介素 1β(IL 1β)和白介素 6 (IL 6 )分泌水平。结果 :(1)慢性应激大鼠的胸腺指数和脾指数减小 ,T淋巴细胞和B淋巴细胞增殖受到抑制 ,IL 1β 和IL 6分泌水平降低 ,提示慢性应激大鼠的免疫功能受到抑制。 (2 )柴地合方治疗可使慢性应激大鼠受抑制的免疫功能得到改善 ,对慢性应激导致的病理性免疫功能抑制具有增强作用 ,而氯丙咪嗪对其没有显著改变。结论 :柴地合方可逆转慢性应激大鼠免疫功能的抑制状态 ,这可能是其抗抑郁作用的机理之一 ,并与氯丙咪嗪作用机理可能不同。Objective:Observed the effect about “Chai Di He Fang”,a traditional Chinese medicine on multifold immunologic function of chronic stress rat,for sake of discussing partial antidepressant mechanism about it.Methods:Chronic stress of depression model was made by alternate mulriple stimulate,and lymphocyte of spleen was cultured in vitro.The spleen index and thymus index were measured by weighing the spleen and thymus.The proliferation of T lymphocyte and B lymphocyte were measured by 3H TdR incorporation assay.The level of IL 1 β and IL 6 were measured by radioimmunoassay.Results:(1)The spleen index and thymus index of chronic stress rats decreased,the proliferation of T lymphocyte and B lymphocyte of chronic stress rats was suppressed,and the level of IL 1 β and IL 6 of chronic stress rats was decreased.The result above showed that immunologic function of chronic stress rats was suppressed.(2)“Chai Di He Fang”improved the pathologic immunosuppression that took place in chronic stress butthe chlorimipramine did not.Conclusion:The results suggest the improvement of pathologic immunosuppression in depression may be one of the mechanisms of antidepressant effect of “Chai Di He Fang” and there must be some difference between the mechanism of antidepressant effect of “Chai Di He Fang” and that of chlorimipramine.
