摘 要:目的 :运用Lowicry1K4M低温包埋及免疫电镜法对甲状腺滤泡细胞源性肿瘤及瘤样病变中甲状腺球蛋白 (Tg)、癌基因蛋白 (Ret)、黏糖蛋白 1(Muc 1)和Galectin 3(Gal 3)基因蛋白产物的表达进行检测。 方法 :对 18例患者手术切除的甲状腺标本进行低温包埋和免疫电镜研究。 结果 :良、恶性甲状腺滤泡上皮细胞的细胞质中胶质小滴、粗面内质网、高尔基复合体及近滤泡的吞饮泡、滤泡腔中有Tg胶体金颗粒 ;本组 9例良性病变 (甲状腺腺瘤和结节性甲状腺肿 )标本Ret、Muc 1和Gal 3三种标记均阴性 ,乳头状癌和滤泡性癌标本的Ret标记可见癌细胞的细胞质中核膜外粗面内质网的核糖体上、粗面内质网腔内及细胞质小泡中有金颗粒的积聚 ,而线粒体、胶质小滴、细胞核、滤泡腔中为阴性 ;Muc 1金颗粒见于粗面内质网、细胞质小泡和细胞膜下 ,细胞核、线粒体、胶质小滴、滤泡腔为阴性 ;Gal 3金颗粒分布较广 ,细胞核常染色质区多见 ,核膜下、粗面内质网、细胞质小泡以及胶质小滴的外周质膜下亦有分布 ,滤泡腔为阴性。 结论 :Ret、Muc 1和GalObjective:The expressions of Tg、Ret、Mucin-1 and Galectin-3 in follicular tumors and tumor-like lesions of thyroid were carried out by using immuno-electron microscopic study with the method of Lowicrry1 K 4M low temperature embedding. Methods:Eighteen cases of thyroid lesions were treated by Lowicry1 K 4M low temperature embedding and Immuno-electron microscopic study. Results: A few clusters of Tg gold particles were found in the cytoplasmic organells such as droplet of colloid, rough endoplasmic reticulum(RER), Golgi complex as well as pinosome of cytoplasm and follicle space in both benign and malignant thyrocytes. There was no any gold particle of Ret, mucin-1 and galectin-3 labelling in the cases of benign lesions such as thyroid adenoma and nodular goiter . In papillary carcinoma and follicular carcinoma, the expression of Ret labelling showed that there were a few clusters of gold particles in ribosome attaching to the RER, the chamber of RER and the vesicle of cytoplasm. The expression of Muc-1 showed the same as the Ret except the distribution of cell membrane, but there was no expression in mitochondrion, droplets of cytoplasm, nucleus or the cavity of follicle found. The distribution of Galection-3 was fairly extensive, with more aggregation in euchromatin of nucleus and RER, the vesicle of cytoplasm and beneath of plasma membrane, but none in the cavity of follicle. Conclusion:Since the different concordance among ultrastructural localization of Ret, Mucin-1, and Galectin-3 discovered in this research the markers of malignant rthyroid tumors may reflect the probable discrepancy in the fields of cell biologic function.
