肺癌细胞株对6种抗癌药物敏感性测定

摘  要:目的 探讨肺癌细胞株抗癌药物的敏感性。方法 采用体外MTT比色分析法对 10株肺癌细胞进行NVB、GEM、TAX、SN - 38、CDDP、5 -Fu 6种抗癌药物的体外敏感试验。结果  ( 1) 6种抗癌药物对SCLC的显效顺序依次为GEM、NVB、CDDP、TAX、5 -Fu、SN - 38,4株SCLC的敏感性顺序依次为Lu135、N2 31、H6 9、SBC - 3;( 2 ) 6种抗癌药物对NSCLC的显效顺序依次为 5 -Fu、NVB、GEM、CDDP、TAX ,对SN - 38均为耐药 ,3种 6株NSCLC的敏感性顺序依次为Lu 6 5、PC 7、LK - 2、PC 9、PC 14、A 5 49;( 3)SCLC株对 6种抗癌药物敏感性优于NSCLC ,6种抗癌药物对 4株SCLC的总有效率为 87 5 % ,敏感为 6 2 5 % ,较敏感为 2 5 % ,耐药为12 5 % ;对 6株NSCLC的总有效率为 5 2 78% ,敏感为 16 6 7% ,较敏感为 36 11% ,耐药为 4 4 4 4%。结论  10株肺癌细胞株对 5 -Fu最敏感 ,GEM、NVB、CDDP、TAX次之 ,SN - 38效果最差。 6种药物中对 5 -Fu、CDDP未表现出明显耐药性 ,其余 4种均存在不同程度的耐药性 ,表明肺癌细胞株存在自然耐药性。Objective To investigate the sensitivity of lung cancer cell lines to anticancer drugs. Methods The sensitivity of 10 lung cancer lines to NVB, GEM, TAX, SN-38, CDDP and 5-Fu was detected by MTT colorimetric assay in vitro. Results a. 6 types of anticancer drugs to which SCLC cell lines responded were NVB, GEM, TAX, SN-38, CDDP and 5-Fu in descending order of response. The 4 sensitive SCLC cell lines in descending order of resistance were Lu135, N231, H69 and SBC-32) six types of anticancer drugs to which NSCLC cell lines responsed were 5-Fu, NVB, GEM, TAX, CDDP and SN-38 in descending order of response. 6 cell lines of three types of NSCLC which responsed to anticancer drugs were Lu65, PC7, LK-2, PC9, PC14 and A549 in descending order of resistance. The sensitivity of SCLC cell lines to 6 types of anticancer drugs was superior to that of NSCLC cell lines. The overall response rate of 4 SCLC cell lines treated with 6 types of anticancer drugs was 87.5%. The high sensitivity rate was 62.5%. The most sensitivity rate was 25%. The resistance rate was 12.5%. The overall response rate of 6 NSCLC was 52.78%. The high sensitivity rate was 16,67%. The most sensitivity rate was 36.11%. The resistance rate was 44.44%. Conclusion Ten lung cancer cell lines were high in sensitivity to 5-Fu, were median sensitive to GEM, NVB, CDDP, TAX, were not sensitive to SN-38. The lung cancer cell lines did not show resistance to 5-Fu and CDDP. But they showed different resistance to the other 4 types of anticancer drugs. We concluded that lung cancer cell lines had intrinsic resistance to anticancer drugs.
