摘  要:观察重组人肿瘤坏死因子α(rh—TNFα)及其与化疗药物合用对五株消化道癌细胞的细胞毒作用,结果发现,rh—TNFα对五种细胞株均有不同程度的细胞毒效应。随着浓度的增高,其作用呈递增趋势。其中尤以对213细胞和C1184细胞的作用为明显(P<0.05);当rh—TNFα与5—Fu或PYM或MMC合用时,其细胞毒作用明显增强(P<0.05)。提示rh—TNFα对几种消化道癌细胞具有较广谱的杀伤活性,对临床试验治疗具有一定的参考价值。Tumor necrosis factor (TNF) is a cytokine with a broad range of biological activities. It has cyto-toxic and/or cytostatic effects on a variety of tumor cells in vitro, but no deleterious effects on normaltissues were seen. the cytotoxic effects of recombinant human tumor necrosis factor--α(rh-TNFα)alone or in combination with 5--Fu, Pinyangmycin(PYM) and mitomycin C (MMC),on five cancercell lines of the gastrointestinal tract were observed. The results showed that the cytotoxic effect of rh--TNFα on the five cancer cell lines was different. With the increase of the concentration of rh--TNFα, its cytotoxic activity increased progressively (P<0. 05),especially on 213 cell and C1184 cell,and the effects of rh--TNFα combined with antitumor drugs were significantly enhanced, as comparedwith rh--TNFα alone. The results confirm that the spectrum of the activity of rh--TNFα on gastroin-testinal cancer cells in vitro is rather broad, particularly combined with antitumor drugs, and that dif-ferent cancer cell line has different sensitivity to rh--TNFα.
