摘 要:本文通过免疫组织化学方法,对石蜡切片的骨巨细胞瘤及体外培养的肿瘤性多核巨细胞进行碳酸酐酶Ⅱ染色,结果发现在石蜡切片中的95%以上多核巨细胞与破骨细胞一样呈强阳性表达,部分(50%左右)圆形单核基质细胞呈阳性或弱阳性表达,梭形单核基质细胞呈阴性。玻片培养的多核巨细胞也是阳性但着色较浅。本研究表明多核巨细胞与破骨细胞性骨吸收有共同之处,都是通过碳酸酐酶产酸后经质子泵分泌到局部微环境中进行脱矿。本研究进一步证实了肿瘤性多核巨细胞可能与破骨细胞一样来源于同一前体细胞。In this study we stained the paraffin sections of giant cell tumour (GCT) of bone and the coverslips of multinucleated giant cells (MGC) in vitro by immunohistochemical method, with anti-carbonic anhydrase Ⅱ (CAⅡ)antibodies. In paraffin sections,over 95% of MGC expressed the CAⅡ,a few of round-shaped stromal cells(about 50%)expressed CA Ⅱ weakly and the spear-shaped stromal cells did not express CA Ⅱ. In coverslips in vitro,MGC also expressed CA Ⅱ but the colour was light. It indicated that the MGC bone resorption,just like that of osteoclasts,required the action of proton pumps to transport acid, which generated from CA Ⅱ,into the local microenviromment to dissolute the mineral. It further demonstrated that MGC and the osteoclasts might be derived from the same precursor cells.
