摘 要:目的探讨2'-O-meRNA通过抑制端粒酶来杀伤胃癌细胞系SGC-7901的作用及对化疗药5-FU和BCNU疗效的影响。方法培养人胃癌细胞系SGC-7901,分为5-FU组、BCNU组、2'-O-meR- NA组、反义对照组、化疗药与反义抑制剂合用组,观察用药后该细胞系存活情况及端粒酶活性。结果5- FU、BCNU、2'-O-meRNA可有效抑制SGC-7901细胞的活性(P<0.05),BCNU、2'-O-meRNA可有效抑制细胞端粒酶活性,1μM 2'-O-meRNA可加强6.25 μg/ml BCNU的疗效(P<0.01),但是不能加强12.5μg/ml 5-FU的作用(P>0.05)。结论2'-O-meRNA抑制胃癌细胞系SGC-7901的生长。1 μM 2'-O- meRNA与6.25 μg/ml BCNU合用时有协同作用,2'-O-meRNA与5-FU合用时无协同作用。Aims To study the effect of 2'-O-mRNA on the susceptibility of human gastric cancer cell line SGC-7901 and telomerase. Methods HTR antisense 2'-O-mRNA with sequence CAGUUAGGGUUAG which can recognize the RNA template of telomerase of SGC-7901 was transfected by lipofectamine, drug sensitivity test was performed with MTT method, and telomerase activity was quantitatively determined by TRAP-PCR-ELISA method. Results Telomerase activity was dramatically declined during the cell incubation with hTR antisense 2'-O-mRN、BCNU plus 2'-O-mRNA, but not 5-FU. Conclusions The hTR antisense 2'-O-mRNA maybe correlates with its inhibitory effects on telomerase activity and the ability of inducing apoptotic cell death, and provides a basis for the development of SGC-7901 associated with BCNU, not with 5-FU. These findings indicate that hTR antisense 2'-O-mRNA may represent a novel potential chemosensitisation.
