摘 要:目的 利用杂交瘤技术制备脐血树突状细胞和食管癌细胞融合疫苗 ,研究融合疫苗的生物学特征。方法 免疫磁珠法分离脐血CD34+ 干细胞 ,细胞因子诱导扩增为成熟树突状细胞 (DC) ;用聚乙二醇法 (PEG)将DC与食管癌细胞EC10 9融合 ,免疫磁珠法筛选阳性克隆EC10 9-DC ;利用流式细胞术鉴定融合疫苗表型 ;绘制融合疫苗生长曲线。结果 融合疫苗EC10 9-DC可在体外培养生长 ,高表达CD80 ,CD83和CD86 ,增殖活性低于EC10 9。结论 融合疫苗EC10 9-DC细胞具备了刺激免疫细胞活化的分子表型 ,为食管癌的特异性免疫治疗提供新的理论依据。Objective To establish the cancer vaccine(EC109-DC)prepared by fusions of esophageal carcinoma cell line with dendritic cell derived from cord blood and to study its biological characteristics. Methods CD34+ hematopoietic stem cells were isolated from cord blood using CD34+ progenitor cell isolation kit by magnetic cell sorting system(MACS); The CD34+ cell were expended with rhGM-CSF, rhTNF-α and rhSCF for 2 weeks as DC, and fallowed by fusion of PEG-3600 with esophageal carcinoma cell line. MACS marked with HLA-DR MicroBeads was used to generate the EC109-DC. The phenotypes and proliferation were evaluated by flow cytometry and technique of cell culture in vitro. Results The EC109-DC cells could proliferate slowly in vitro and highly expressed CE80, CD83 and CD86. Conclusion EC109-DC vaccine obtained by PEG fusion acquire the immuno-stimulating phenotype. The results of this research provide the new means for the DC based vaccine against esophageal carcinoma.
