摘 要:目的 比较艾蒿花粉和豚草花粉粗浸液中的蛋白质组分 ,探讨两者间有无共同抗原。方法 制备艾蒿花粉和豚草花粉的花粉浸液 ,采用SDS -PAGE及考马斯亮蓝染色分析两种花粉粗浸液中的蛋白质组分 ;用蒿属花粉过敏者的血清作为第一抗体 ,分别与两种花粉浸液发生反应 ,通过Western -blotting判断两种花粉间交叉抗原的存在。结果 SDS -PAGE显示 ,艾蒿花粉显带 13条 ,分子量在 18~ 10 0kDa之间 ;豚草花粉显带 5条 ,分子量在 18~ 6 3kDa之间。免疫印迹表明 ,11份蒿属花粉过敏患者的血清 ,均能同时与艾蒿花粉及豚草花粉抗原发生反应 ,其中 ,同时发生反应的艾蒿花粉抗原有两条 ,分子量分别为 18kDa和 2 0kDa ;豚草花粉抗原有一条 ,分子量为18kDa。结论 艾蒿花粉与豚草花粉提取液蛋白组分差异较大 ,前者更复杂 ;但两者间存在共同抗原 ,可发生交叉过敏。Objective To compare proteins in the extract of Artemisia pollen and ragweed pollen and to examine the existance of common antigen between the two types of pollens. Methods Extract of artemisia pollen and ragweed pollen were prepared SDS-PAGE and Coomassis were used to analyze proteins. Sera from patients with artemisia pollergy was used as the first antibody and Western-blotting was used to examine the existance of cross-reactive antigens. Results SDS-PAGE suggested that artemisia pollen displayed 13 bands and the range of molecular weight was 18~100 kDa. Ragweed pollen displayed 5 bands and the range of molecular weight was 18~63 kDa. West-blotting indicated that the extract of artemisia pollen had two bands that reacted with IgE in the sera from patients with Artemisia pollen allergy,and, the molecular weight of the two bands was 18 kDa and 20 kDa, respectively. The extract of ragweed pollen had 1 bands that reacted with the same sera and its molecular weight was 18 kDa. Conclusion Significant difference exists between the extract of artemisia pollen and ragweed pollen, with the former being more complicated. However, commom antigens exist and cross-reactivity can occur.
