人胃粘液腺癌裸鼠移植瘤模型的建立及其生物学特性的研究

摘　　要：将手术切除的原发性人胃癌新鲜组织移植于裸鼠皮下获得成功,并已经传至15代。建立的人胃粘液腺癌裸鼠移植瘤模型(GC—916)具有移植成功率高,带瘤时间长的特点。总移植成活率99％,移植瘤生长潜伏期14天,裸鼠最长带瘤存活时间为160天,平均带瘤存活时间为89天,经组织学、组织化学、免疫病理学和超微结构观察,证实裸鼠移植瘤保持了原发人体肿瘤的结构和功能,移植传代的胃癌细胞具有表达突变型P_(53)蛋白及产生癌胚抗原的特性。该移植瘤模型的建立为进一步研究人胃癌的基础理论和临床治疗提供了较理想的实验动物模型。A human primary gastric mucoid adenocarcinoma from a surgical specimen was successfully transplanted into nude mice and has been transferred through 15 passages in 80 nude mice with the transplantable rate of 99%. The transplanted tumor is designated as transplantable human primary gastric cancer-916 in nude mice(GC-916),The latent period of the transplanted tumor was 14 days. The average tumorbearing life time of mice was 89 days. Histology, histochemistry, immunopathology and ultrastructre showed that the transplantable tumor retained the original structures and functions of the human primary gastric carcinoma. It demonstrated that the transplantable tumor possessed the features of expressing mutant p53 proteins and producing carcinoembryonic antigens. This animal model could be useful in the study of biological behavior of human gastric carcinoma and provide an important tool for experimental therapy.
