摘 要:目的在体外研究视黄酸(retinoicacid,RA)对小鸡后极部巩膜成纤维细胞MMP-2表达的调节作用,探讨RA在形觉剥夺性近视(form-deprivationmyopia,FDM)中的作用。方法取40只1d龄来亨雏鸡以半透明眼罩遮盖右眼14d制备FDM动物模型,未遮盖眼为正常眼。将正常小鸡眼后极部巩膜成纤维细胞作体外培养并传2代细胞作为正常对照组细胞,FDM眼小鸡后极部巩膜成纤维细胞作体外培养并传2代细胞作为FDM组细胞,取其中20只正常小鸡眼第2代后极部巩膜成纤维细胞,加入不同终浓度RA(6×10-7,6×10-6,6×10-5和6×10-4mol/L)培养24h作为RA组细胞。各组细胞均采用SABC免疫组织化学染色法检测MMP-2的表达,进行计算机图像分析及统计学检验。结果MMP-2在培养的正常鸡巩膜成纤维细胞胞质呈弱阳性表达。FDM组鸡巩膜成纤维细胞较对照组MMP-2表达明显增高(P<0.01)。6×10-7mol/LRA组与对照组比较无统计学意义(P>0.05)。6×10-6和6×10-5mol/LRA组较对照组MMP-2表达增高(P<0.01),6×10-4mol/LRA组细胞普遍变形崩解。各视黄酸浓度组之间差异有统计学意义(P<0.01)。结论在一定浓度范围内,RA上调体外培养的鸡巩膜成纤维细胞MMP-2的表达。RA可能是调节巩膜MMP-2表达的重要因素而参与FDM形成。AIM: To study the effects of retinoic acid (RA) on the expression of matrix metalloproteinases-2 (MMP-2) in chick posterior scleral fibroblasts (CPSFs) in vitro and to clarify the role of RA in the mechanism of FDM.· METHODS: The right eyes of 40 1-day-old leghorn chicks were covered with a plastic goggle to induce FDM for 14d after birth, with the left eyes as the control. CPSFs of each group were transferred of culture in vitro . Meanwhile, early passage (second passage) of 20 normal age-matched CPSFs were incubated for 24h with different final concentrations of RA (6×10-7, 6×10-6, 6×10-5 and 6×10-4mol/L). MMP-2 expression in CPSFs was evaluated respectively by immunohistochemistry method. The results were analyzed with computer image analysis system and statistical test.· RESULTS: The cultured CPSFs expressed MMP-2. The difference was not statistical significant between CPSFs incubated with 6×10-7mol/L RA and the controls (P > 0.05). In comparison with the controls, the MMP-2 expression of FDM Group were much higher (P <0.01). The MMP-2 expression of CPSFs incubated with 6×10-6 and 6×10-5mol/L of RA was higher than Control Group (P <0.01). CPSFs incubated with 6×10-4mol/L of RA came to degeneration and death.· CONCLUSIONS: RA can up-regulate the expression of MMP-2 in CPSFs and appears to play an important role in the mechanism of FDM.·
