人参单体皂甙Rh2对人喉癌细胞株Hep-2抗增殖作用的研究

摘　　要：目的　探讨人参单体皂甙Rh2对体外培养的人喉癌细胞株Hep - 2的抗增殖作用及其作用机制。方法　采用四甲基偶氮唑蓝 (MTT)实验、流式细胞技术及免疫组织化学S -ABC法分别检测人参单体皂甙Rh2对喉癌细胞株Hep - 2的增殖、细胞周期的影响。 结果　 (1)人参单体皂甙Rh2可明显抑制Hep - 2细胞的生长 ,并呈剂量、时间依赖性作用。 (2 )人参单体皂甙Rh2 (30 μg/ml)处理细胞 2 4h :G1期细胞由 6 0 0 9%增加至6 8 86 % (P <0 0 1) ,S期细胞由 18 89%减少至 12 30 % (P <0 0 1) ,G2 /M期细胞及凋亡细胞无明显变化 (P>0 0 5 ) ,细胞阻滞于G1期。结论　人参单体皂甙Rh2对人喉癌细胞株Hep - 2具有明显的生长抑制作用 ,并可导致其G1期细胞周期阻滞。Objective To study the anti-proliferative effect and the mechanism of Ginsoniside Rh2(G-Rh2) on human laryngeal carcinoma cell line Hep-2.Methods MTT assay was used to evaluate the growth inhibition effect of G-Rh2 on human laryngeal carcinoma cell line Hep-2 and the flow cytometry for cell cycle analisis.Results G-Rh2 significantly inhibited the growth of Hep-2 cells with a dose-time-independent manner. After 24 hours treatment with 30μg/ml G-Rh2, the proportion of cells in G 1 phase increased from 60.09% to 68.86%(P<0.01), while the cells in S phase decreased from 18.89% to 12.30%(P<0.01) and there is no significantly change about cells in G 2/M phase and apoptotic cells.Conclusions G-Rh2 inhibited the growth of human laryngeal carcinoma cell line Hep-2 significantly, arrested cell cycle in G 1 phase. It is the anti-tumor mechanism for G-Rh2 possibly by arresting cell cycle.
