VEGF-C和VEGR-3在舌癌组织中的表达及其和肿瘤淋巴转移的关系

摘　　要：目的　检测血管内皮生长因子C(VEGF -C)和它的受体VEGFR - 3在舌癌组织中的表达。方法　采用免疫组化法检测 6 2例舌癌组织中VEGF -C和VEGFR - 3的表达及定位。结果　 6 2例舌癌中 ,2 4例VEGF-C染色阳性 ,阳性 38.7%。且发现肿瘤细胞中VEGF -C的表达和内皮细胞中VEGFR - 3的表达有显著相关性 ,形态学研究证实 ,此内皮细胞为淋巴管内皮细胞 ,这些淋巴管主要位于肿瘤周围 ,在肿瘤和正常组织交界区即肿瘤浸润区表达尤其丰富。临床病理研究证实 ,VEGF -C的表达和肿瘤淋巴转移密切相关。结论　VEGF -C主要由肿瘤细胞产生 ,VEGF -C可能通过表达在淋巴管内皮细胞上的VEGFR - 3来诱导舌癌间质淋巴管的增生。在这样的微环境下 ,肿瘤细胞通过增加的肿瘤细胞和淋巴管接触点而能容易地侵入淋巴管 ,到达淋巴结 ,并在那增值 。Objective To investigate the expression of vascular endotheliar growth factor-c(VEGF-C) and it's receptor VEGFR-3 in human tongue cancer.Methods Paraffin-embedded specimens from 62 patients with tongue cancer were examined by immunohistochemical staining.Results Positive immunohistochemical staining for VEGF-C was observed in 24 of all patients the positive ration was 38.7%.A significant positive correlation was found between VEGF-C in cancer cells and VEGFR-3 in vascular endothelial cells.Morphologically VEGFR-3 expression was detected in the endothelial cells of the stromal lymphatic vessels which located in the bound also called the cancer intravation region between the tumour islands and normal tissue.In this cohort of tongue cancer patients,VEGF-C expression was significantly associated with lymph node metastasis.Conclusion VEGF-C was mainly produced from cancer cells.These results strongly indicate that VEGF-C may induce the proliferation of lymphatic vessels in the stroma of tongue cancer via activiation of VEGFR-3,expressed on the endothelial cells of lymphatic vessels.In these circurnstances,cancer cells can easily invade the lymphatic vessels,because of the increase of the contact points of cancer cells with the lymphatic vessels.Then,cancer cells migrate into the regional lymph nodes,and proliferate there to form a metastasis.
