摘  要:目的 :探讨丹参对过氧化氢 (H2 O2 )诱导的大鼠主动脉平滑肌细胞 (VSMCs)凋亡的影响及其可能机制。方法 :选用体外培养的第 3~ 5代大鼠VSMCs,以H2 O2 作为外源性活性氧 (ROS)诱导VSMC凋亡 ,用丹参进行干预 ,以MTT比色法测定细胞存活率 ,流式细胞术检测细胞凋亡率 ,同时用免疫细胞化学法检测VSMCs 中Bcl 2和Bax蛋白表达的变化。结果 :1mmol/LH2 O2 诱导VSMCs 凋亡 ,细胞存活率明显低于对照组 ,细胞凋亡率、Bax/Bcl 2蛋白表达的阳性指数 (PI)明显高于对照组 (P <0 0 1) ;丹参干预后 ,和H2 O2 组相比 ,细胞存活率明显升高 ,而细胞凋亡率、Bax/Bcl 2蛋白表达的PI显著降低 (P <0 0 5或P <0 0 1)。结论 :丹参可通过下调Bax/Bcl 2蛋白表达而对抗H2 O2 诱导的VSMCs 凋亡。Objective: To investigate the effects of Salviae miltiorrhizae on H 2O 2 induced apoptosis in rat vascular smooth muscle cells(VSMC s) and its related mechanism Methods: The 3~5 passages of VSMC s were used, H 2O 2 was added to the medium of cultured rat aortic VSMC s as extraneous reactive oxygen species(ROS) The viability percentage of VSMC s was evaluated by MTT colorimetric assay. The percentage of apoptosis was measured by flow cytometry. The expression of Bax/Bcl 2 protein was examined with immunohistochemical technique and positive index(PI) was calculated Results: 1mmol/L H 2O 2 could increase apoptosis rate. The PI of Bax/Bcl 2 protein in H 2O 2 group was higher(P<0 01) When VSMC s were incubated with H 2O 2 plus Salviae miltiorrhizae , the apoptosis rate greatly declined, and at the same time, the PI of Bax/Bcl 2 protein reduced remarkalby compared with H 2O 2 group (P<0 05 or P<0 01) Conclusion: Salviae miltiorrhizae could prevent VSMC s from H 2O 2 induced apoptosis by decreasing the Bax/Bcl 2 protein expression
