微卫星不稳定性在胃癌组织中的表现及意义

摘　　要：目的　研究胃癌微卫星不稳定性 (MSI)的情况及其在胃癌发生发展中的意义。方法　应用显微切割技术、聚合酶链式反应、变性聚丙烯酰胺凝胶电泳、银染等方法 ,分别对 17例乳头状及管状腺癌、19例低分化腺癌和 16例黏液腺癌的石蜡包埋组织MSI的表现情况进行研究。结果　各型胃癌MSI阳性率明显高于轻度慢性浅表性胃炎组 (P <0 .0 5 ) ,乳头状及管状腺癌组MSI阳性率显著高于低分化腺癌、黏液腺癌组 (P <0 .0 5 )。结论　MSI在胃癌发展中有一定作用 ,参与了胃癌的发展分化过程 。Objective To study the expressions and significances of microsatellite instability (MSI) in gastric carcinoma.Methods The expression of MSI were examined by using microdissection polymerase chain reaction(PCR),denatured-polyacrylamide gel-electrophoresis and silver staining in paraffin section of 52 cases of gastric carcinoma,including 17 cases of papillary and canalicular adensma,19 cases of poorly differentiated edenorna and 16 cases of mucinous adenocarcinoma.Results The rates of MSI in three typies of gastric carcinoma were markedly higher than that in chronic gastritis.The rates of MSI in gastric papillary and tubular adenocarcinoma was markedly higher than that in gastric poorly differentiated adenocarcinoma and mucoid adenocarcinoma.Conclusion The MSI play a part in not only the development of gastric carcinoma but also differentiated stage of gastric carcinoma,and gastic carcinoma with higher positive rate of SMI possesses lower degree of malignancy.
