摘 要:目的 :研究在上皮性卵巢癌细胞株中卵泡刺激素 (FSH)受体的不同表达并通过不同浓度的FSH作用 ,观察FSH刺激后各卵巢癌细胞株的增殖与上皮钙粘素 (E -cadherin)在各细胞株中的表达情况。方法 :应用逆转录聚合酶链反应(RT -PCR)及免疫细胞化学方法研究体外培养上皮性卵巢癌细胞株HO - 8910、HO - 8910PM及SKOV3中FSH受体表达 ,免疫组化半定量研究各细胞株中E -cadherin表达及其在FSH作用后的表达情况 ,噻唑蓝染色法了解FSH作用后各细胞株的细胞增殖程度。结果 :在卵巢癌细胞株HO - 8910及HO - 8910PM中存在着FSH受体 ,SKOV3细胞株中的FSH受体表达阴性。用不同浓度FSH培养 4 8小时后 ,HO - 8910及HO - 8910PM的细胞增殖指数高于SKOV3。当FSH浓度为 4 0U/L时细胞增殖指数最高 (P <0 .0 5 )。E -cadherin在 3株细胞中的表达随试验中细胞株转移程度的不同而具有差异 ,FSH培养后 ,FSH受体阳性细胞株HO - 8910及HO - 8910PM的E -cadherin表达均减弱。结论 :FSH是FSH受体阳性卵巢癌细胞增长的促进因素并可引起受体阳性肿瘤细胞E -cadherin表达的下调 ,从而增强卵巢癌细胞的转移能力。Objective: The expression of follicle stim ulating hor mone (FSH) receptor in human epi thelial ovarian c ancer cell lines was investigated and the effects of diffe rent levels of FSH on cell proliferation and on expression of epithel ial cadherin (E-ca dherin) of these ovarian cancer cell lines were examined dlso.Methods: The FSH receptor expression were d etected in ovarian caheer cell line HO- 8910,HO-8910PM,and SKOV3 by reverse tran scription poly merase chain reaction (RT-PCR) and immunohi stochemical (IHC) mechod cell prolifera tion were detected by methyl thia zolyl tetrazolium (MTT); the expression of E-cadherin were detected by IHC.Results: P ositive FSH receptor mRNA expression was fo und by RT-PCR in HO-8910 and HO-8910P M, FSH receptor expression in SKOV3 was neg ative; IHC results was similar to RFPCR. A do se-response effect was seen with FSH in cell proliferation of HO-8910PM and HO-8910, wit h a maximally stimulating FSH conce ntration of 40 U/L for a period of 48 hour. This effect was not observed in SKOV3 . The expression of E-cadherin was we aker in HO-8910PM than HO-8910 and SKOV3. Af ter stimulated by FSH, the E-cadheri n expression was decreased in HO-8910 and HO -8910PM, and no change in SKOV3. Conclusions: FSH is a promoting factor in proli feration of prositive FSH receptor ovarian cancer cell l ines and can down-regulate the E- cadherin expression level, theresore to r educe the intercellutar adhesion of positi ve FSH recepto r ovarian cancer cell lines, and to enhenc e the ability of cancer cell metastasis.
