血管抑素对人胰腺癌细胞生长的体内抑制作用

摘　　要：目的　探讨血管抑素 (An)对实验性人胰腺癌的治疗作用及生物学特征。方法　用人胰腺癌细胞系Patu 8988接种裸鼠制备动物模型 ,用An(2 5、5 0、10 0mg/kg)对瘤部进行注射治疗 30天 ,观察动物行为 ,测量不同治疗剂量组与对照组瘤体大小 ;对肺、肝、肾及肿瘤进行病理学检查 ,并对其肿瘤区的血管密度进行检测。结果　在肿瘤种植 30天时 ,对照组与 2 5、5 0、10 0mg/kgAn组肿瘤重量分别为 1 77± 0 4 2、0 74± 0 4 0、0 4 9± 0 2 1和 0 2 5± 0 13g ,An抑瘤率分别为 5 7%、72 %和 86 % ,10 0mg/kgAn抑瘤作用最明显 ,此外瘤体体积、微血管形成也有相应变化。结论　An对胰腺癌生长有明显的抑制作用 ,可作为临床上一种新的抗肿瘤药物。Objective To observe the therapeutic effect of angiostatin (An)in human pancreatic cancer cell growth.Methods Human pancreatic cancer cell line Patu 8988 was planted in the naked mice,and was injected into the animal models in 0,25,50 and 100 mg/kg respectively for 30 days.Then the weight and the volumes of the cancer tissue was measured,pathologic examination was done in the lungs,livers and kidneys of the animal models,and the dentisy of blood vessels in cancer tissue was observed.Results The weight of animal models with An in 0,25,50and 100 mg/kg were 1.77±0.42,0.74±0.40,0.49±0.21 and 0.25±0.13 g,respectively and the inhibition rate of cancer tissue growth by the An were 57%,72% and 86% respectively.Additionally there was obvious variation of cancer tissue volume between different groups of An therapy.Conclusion An could inhibit the growth of pancreatic cancer tissue in animal models,suggesting it could be applied in cancer therapy in clinic.
