摘  要:目的 探讨自拟中药方———脑复聪颗粒对脑缺血小鼠记忆能力及大脑皮层超氧化物歧化酶 (SOD)和谷胱甘肽过氧化物酶 (GSH -Px)活性的影响。方法 采用双颈总动脉闭塞 -缺血 -再灌术造成学习记忆障碍小鼠模型。小鼠记忆能力以Morris圆形水迷宫的潜伏期作为指标。结果 小鼠脑缺血后 7d潜伏期〔(82 3± 3 2 0 )s〕比假手术组〔 (3 5 3± 3 7 9)s〕延长 (P <0 0 1) ;脑复聪组〔 (3 0 6± 3 1 3 )s〕恢复正常 (P <0 0 1)。脑缺血小鼠大脑皮层SOD和GSH -Px活性 (68 5 9± 2 5 5 ,10 88± 1 0 1)比假手术组 (79 68± 5 69,13 0 9± 2 5 3 )降低 ,脑复聪及当归芍药散治疗组提高到正常水平 (77 5 6± 4 88,13 3 5± 3 3 8;75 77± 6 88,13 3 2± 2 69) (P <0 0 5~ 0 0 1)。结论 脑复聪颗粒对脑缺血所致的智能障碍有保护作用 ,其机制可能与提高SOD和GSH -Px活性、对抗自由基损伤有关。Objective To investigate the effects of Naofucong grain on memory and cerebral superoxide Dismutase(SOD)、glutathione perioxidase(GSH-Px)activities in cerebral ischemic mice. Methods Mice models of learning/memory impairment were induced by bilateral ligation of the internal carotid arteries.Memory abilities were assessed by measurements of latent period in the Morris Water Maze. Results On the 7th day after the operation of bilateral ligation,latent periods〔(82.3±32.0)s〕were longer than those of pseudo operated mice〔(35.3±37.9)s〕(P<0.01);administration with Naofucong grain ameliorated those changes〔(30.6±31.3)s〕(P<0.01).The SOD and GSH-Px activities in brain cortex(68.59±2.55,10.88±1.01)were lower than those in pseudo operated mice(79.68±5.69,13.09±2.53),administration with Naofucong grain or Dangguishaoyao powder showed amelioration to normal levels(77.56±4.88,13.35±3.38;75.77±6.88,13.32±2.69)(P<0.05~0.01) Conclusion Naofucong grain possesses protective function on intellectual deficits in cerebral ischemic mice,which may be related to the enhancement of the cerebral SOD、GSH-Px activities.
