摘 要:通过电镜观察附着于骨片的骨巨细胞瘤多核巨细胞,表明在骨吸收时具有褶皱缘结构。采用可标记破骨细胞褶皱缘的空泡型质子泵31KD亚基的抗体,对体外培养的骨巨细胞瘤细胞进行免疫组织化学染色,结果显示多核巨细胞为阳性,间质细胞为阴性,阳性区主要分布于多核巨细胞浆内的空泡区及伪足区。通过mRNA原位杂交方法检测到,多核巨细胞和间质细胞中均有空泡型质子泵的3lKD亚基mRNA表达。结果提示多核巨细胞与破骨细胞可能有相似的骨吸收泌酸机制。Expression of V-ATPase in human osteoclast-like cells (MGC) from osteoclastoma (GCT) was detected to compare the mechanism of bone resportion between the human osteoclast-like cells from the osteoclastoma and the osteoclasts. Methods:Transmission electron microscopy4in situ hybridization and immunocytochemistry were used. Results: (1)By transmission electron microscopy we have observed that in the state of bone resorption the human osteoclast-like cells from the osteoclastoma have the construction for secreting acid ruffled borders. (2)With the antibody to V-ATPase 31KD subunit which can labeled the ruffled borders of the osteoclasts,we have detected that MGC are stained positively while the stromal cells are stained negatively by imtnunocytochemistry. The vacuolar areas and the pseudopods are stained darker. (3)By the method of in situ hybridzation we have detected that both MGC and the stromal cells express the V-ATPase 31KD subunit mRNA. Conclusions: MGC and the osteoclasts have the same construction for secreting acid ruffled borders and express the same acid sucreting apparatus of V-ATPase. So we suggest that MGC may have the same acid secreting mechanism.
