嗜酸乳杆菌对小鼠U_(14)移植瘤的抑瘤及凋亡诱导作用的研究

摘　　要：目的　探讨嗜酸乳杆菌对小鼠宫颈鳞癌U14 移植瘤的抑瘤及细胞凋亡的诱导作用。方法　建立 6 1 5近交系小鼠U14 移植瘤动物模型 ,随机分组 ,检测各组的抑瘤率 ;应用HE染色法 ,观察小鼠U14 移植瘤组织的病理组织学特征 ;应用末端脱氧核糖核苷酸转移酶介导的缺口末端标记法检测细胞凋亡指数AI。结果　与对照组比较 ,LA对小鼠U14 移植瘤具有非常显著抑瘤作用 (P <0 .0 1 ) ;病理组织学观察 ,LA组移植瘤组织癌细胞周围有大量淋巴细胞和浆细胞浸润 ,对照组及顺铂组的癌细胞周围未见明显淋巴细胞和浆细胞浸润 ;对照组小鼠与LA及顺铂组小鼠之间 ,AI的差异具有非常显著性 (P <0 .0 1 )。结论　嗜酸乳杆菌对小鼠U14 移植瘤具有显著的抑瘤作用 ,诱导肿瘤细胞凋亡是其抑瘤机理之一。Objective To explore the growth-inhibition and the apoptosis induced by Lactobacillus acidophilus (LA) on murine cervical carcinoma U 14 .Methods The 615-strain mice with U 14 were randomly divided into three groups.The inhibitory rate of each group were detected. The tumor pathologic histology of each group were examined by HE stain.The apoptosis index was assessed by TUNEL.Results Lactobacillus acidophilus had greatly significant antitumor effect on 615-strain mice bearing U 14 (P<0.01); The transplanted tumors of LA group were infiltrated with many lymphocytes and plasma cells,however, in those of DDP and control group there were few lymphocytes and plasma cells showed.M arked difference was found in AI between LA and DDP,LA and control, DDP and control.Conclusion Lactobacillus acidophilus showed a marked inhibition effect on occurrence and development of U14,and its antitumor mechanism may be associated with inducing apoptosis.
