摘  要:目的:观察肺腺癌细胞株在连续化疗过程中erbB-2与c-myc基因的动态变化。方法:人肺癌细胞株经阿霉素、足叶乙甙、顺铂单药及联合用药连续刺激,每种药物分高、低个剂量梯度组,同时设对照组。记录细胞每次刺2激间隔的时间。应用流式细胞术分别检测erbB-2,c-myc的阳性细胞数及平均荧光强度,以此推算出不同药物在不同浓度下连续两三次作用细胞株后以上各蛋白表达的细胞数、平均表达量、总表达量。结果:随着培养时间的延长各项检测指标其阳性细胞的百分数、平均荧光强度及总荧光强度均呈下降趋势。erbB-2在高剂量各组基本上均随刺激次数的增加呈下降趋势,总表达量由94.8~1708.3下降到5.3~81.3;而低剂量的顺铂单药及联合用药组则上升。c-myc则无一定规律。结论:第次化疗后,erbB-2表达增高,随化疗次数增加表达大幅度下1降;化疗剂量大小与c-myc的表达无关。AIM:To observe the dynamic changes of erbB 2 and c myc gene of lung cancer line during continuous chemotherapy. METHODS:The human lung cancer cell line(LPET α 1) was repeatedly treated with anticancer drugs doxorubicin,etoposide,cisplatin and combination of the three drugs,and every drug was given in two concentrations.Control group was set up.The interval of the treatment was recorded.The positive cell number and mean fluorescent intension of erbB 2 and c myc were tested with flow cytometry.The number of cells expressing each protein,the mean quantity of each protein and the total one were calculated after each treatment by different drugs at different concentrations twice or three times. RESULTS:The levels of proteins decreased along with the time of cell culture prolonging, including positive cell number,mean fluorescent intension and the whole fluorescent intension.In high dose groups,the expression of erbB 2 was decreased from 94.8-1 708.3 to 5.3-81.3 with the increased times of treatments,but increased in the lowdosage group of cisplatin and combination group.There was no certain regularity for the expression of c myc. CONCLUSION:The expression of erbB 2 increased after the first chemotherapy,and it was greatly decreased along with the increased times of chemotherapy.There is no correlation between the doses of chemotherapy and the expression of c myc.
