摘  要:目的 观察钙 /钙调素依赖蛋白激酶Ⅱ α(calcium/calmodulin dependentproteinkinaseⅡ α ,CaMKⅡ α)在Alzheimer(AD)患者海马的表达及其与过度磷酸化tau蛋白的关系。 方法 利用免疫组织化学和体视学方法定量分析 10例女性AD患者及年龄、性别等与之匹配的 10例老年非痴呆患者 (对照组 )海马不同区域CaMKⅡ α神经元数目和染色强度的差异 ;同时利用免疫荧光双标方法分析AD患者海马CA1区CaMKⅡ α与过度磷酸化tau蛋白共存神经元的比例。 结果 AD患者海马CA1区CaMKⅡ α阳性神经元数目显著减少 ,残存神经元免疫反应性显著增强 ;此区CaMKⅡ α和过度磷酸化tau蛋白共存比例低 ,仅 32 %过度磷酸化tau蛋白神经元同时表达CaMKⅡ α。 结论 CaMKⅡ α的表达选择性在AD患者海马CA1区受损。CaMKⅡ α与过度磷酸化tau蛋白共存比例提示CaMKⅡ α可能部分参与tau蛋白过度磷酸化。Objective To investigate the expression of calcium/calmodulin-dependent protein kinaseⅡ-α(CaMKⅡ-α) and the relationship with hyperphosphorylated tau protein in the hippocampus of Alzheimer (AD) patients. Methods Using immunohistochemical and stereological methods,it was quantitatively analyzed that the number and optical density of CaMKⅡ-α expressing neurons in the CA1-CA4 hippocampal subfields of 10 female AD patients and 10 non-dementiated female age- and sex-matched controls. It was also observed that the ratio of the neurons colocalizing CaMK Ⅱ-α and hyperphosphorylated tau to the neurons expressing hyperphosphorylated tau in the hippocampal CA1 subfields of AD patients using double-labeling immunoflurenscence methods. Results CaMKⅡ-α containing neurons were selectively lost in the CA1 subfield of AD patients accompanied with the enhanced immunoreactivity of remaining neurons. Furthermore,only 32% hyperphosphorylated tau containing neurons recognized by monoclonal antibody AT-8 were immunoreactive for CaMKⅡ-α. Conclusions The CaMKⅡ-α expression in CA1 neurons of AD hippocampus is vulnerable. The percentage of the colocalization of the CaMKⅡ-α and hyperphosphorylated tau implied that CaMKⅡ-α may partly involve in the hyperphosphorylation of the tau protein.
