摘　　要：目的 :研究丁酸钠对前列腺癌 PC- 3M细胞株的增殖抑制和诱导凋亡作用。方法 :前列腺癌 PC- 3M细胞经不同剂量丁酸钠作用后 ,相差显微镜观察细胞形态变化 ,MTT测定丁酸钠对 PC- 3M细胞的增殖抑制率 ,吖啶橙 /溴化乙锭染色检测凋亡细胞 ,流式细胞仪检测细胞周期及凋亡峰。结果 :丁酸钠对 PC- 3M细胞有显著的增殖抑制作用 ,呈时间剂量依赖性 ;并使 PC- 3M细胞产生凋亡形态学变化 ;细胞周期被阻滞于 G1 / G0 期和 G2 / M期 ,S期细胞减少 ,有亚 G1 峰出现。结论 :丁酸钠对 PC- 3M细胞具有抑制增殖和诱导凋亡作用。Objective To investigate the effects of sodium butyrate on cell growth inhibition and apoptosis in PC-3M cell lines. Methods Sodium butyrate was added to the culture media of PC-3M cells for different hours and then the cell changes were observed by phase-contrast microscope, the inhibitory rate of cell growth was detected by MTT assay, the apoptosis rate was determined by AO/EB assay, the apoptosis peak and the changes of cell cycle were measured by flow cytometry (FCM). Results Cell growth was significantly inhibited by sodium butyrate and the inhibitory effect was in time- and dose-dependent manner. The morphological changes of apoptosis in PC-3M cells were found. The cell cycle was arrested to G 1/G 0 and G 2/M, the number of cells in S phase reduced, sub-G 1 appeared. Conclusion Sodium butyrate can inhibit the cell growth and induce apoptosis in PC-3M.
