摘  要:目的 :探讨马桑内酯 (CL)对分离培养的小胶质细胞的生存和增殖的影响。方法 :采用振摇结合贴壁分离法获得纯化的小胶质细胞 ,MTT比色法观察了CL对培养的小胶质细胞增殖影响的量效 曲线、细胞的存活能力以及小胶质细胞刺激后的形态学变化。结果 :经凝集素细胞化学鉴定 ,分离培养的小胶质细胞纯度达 95 %以上 ;以CL( 1 0 - 5mol/L)可显著诱导小胶质细胞的增殖 ,小胶质细胞的存活率达 99% ;刺激后 ,以反应性小胶质细胞为主。当CL >1 0 - 5mol/L时 ,细胞存活率 <85 %。结论 :CL在一定剂量范围内具有促进小胶质细胞增殖作用 ,可能伴随着细胞功能的活跃 ;Objective: To explore the effect of coriaria lactone (CL) on the survival and proliferation of the isolated microglial cells from newborn rat cerebral cortex. Methods: Vibration and differential adhesion were used for isolation of microglia cells. The survival of microglial cells was examined by the MTT viability assay. Results: Identified by lectin cytochemistry, the purity of isolated microglial cells was over 95%. CL (10 -5mol/L) significantly accelerated the proliferation of microglial cells, and the viability of microglial cells was 99%. The main cellular element of microglia was reactive microglia after stimulation of CL. Once the concentration of CL was over 10 -5mol/L, it was toxic to the cultured microglia. Conclusion: Our results indicate that within range of 10 -5mol/L, CL promotes the proliferation of microglia and activates the cells;when CL concentration is over 10 -5mol/L,toxicity become apparent.
