摘  要:目的 观察维生素E琥珀酸酯 (VES)对雄激素非依赖型人前列腺癌细胞系PC 3 ,及其鸡胚绒毛膜尿囊膜 (CAM )移植瘤生长抑制和凋亡作用 ,探讨VES化学预防前列腺癌的可能机制。方法 应用显微荧光术及流式细胞术 ,检测VES对PC 3细胞的生长抑制和诱导凋亡作用 ;采用免疫组化SP方法及Tunel染色 ,并计算VES对PC 3细胞接种CAM移植瘤的增殖细胞核抗原指数 (PI)和细胞凋亡指数 (AI)。结果 VES对前列腺癌PC 3细胞有生长抑制和诱导凋亡作用 ,随VES浓度增高 ,诱导PC 3细胞凋亡率上升 ,提示VES诱导细胞凋亡呈剂量依赖关系。VES 2 0mg/L作用 48h ,诱导细胞凋亡率为 10 .5 % ;VES诱导PC 3的CAM移植瘤PI减小、AI增大 ,PI与AI呈负相关 (γ =-0 .974,P <0 .0 5 )。结论 VES对PC 3细胞及其CAM移植瘤具有抑制肿瘤细胞增殖、促进细胞凋亡 ,阻滞细胞周期 (G0 /G1期 )进展等作用 ,对前列腺癌具有潜在的化学预防作用。Objective To observe the inhibitory effect of vitamin E succinate(VES) on the growth of androgen-independent prostate cancer cell line PC-3 and the chick chorioallantoic membrane chorioallantoic membrane(CAM) xenograft tumor in vitro.We also investigate the possible mechanisim of chemical prevention effect of VES on the prostate cancer.Methods The growth inhibitory effect of VES and apoptosis was detected by flow-cytometry and fluorescence microscopy.The PCNA index(PI) and the apoptosis index(AI) of VES-treated CAM xenograft tumor was tested by the immunohistochemistry and the Tunel assay.Results VES had an inhibitory effect on the proliferation of PC-3 cell line and the CAM xenograft tumor.It could also induce the apoptosis in a dose-dependent manner.The apoptosis percentage was about 10.5% when treated with VES 20 mg/L for 48hr.PI of the CAM exnograft tumor deceased and AI increased after the treatment of VES,showing a negative correlation(γ=-0.974,P<0.05).Conclusion The possible mechanisms of VES inhibitory effect on the prostate cancer include the cell cycle G_0/G_1 arrest,anti-proliferation and apoptosis induction.It suggests that VES has potential chemical prevention effect on the prostate cancer.
