摘  要:目的 :研究骨髓间充质干细胞 (MSC)对大鼠脑缺血再灌注损伤的保护机制。方法 :192只SD大鼠随机分为假手术组、手术组、对照组和MSC治疗组。MSC移植前应用BrdU标记 ,再灌注 1d后经同侧颈内动脉注射MSC(1× 10 6) ,对照组则注射PBS。采用线栓法建立脑缺血再灌注模型。术后每天应用爬杆法评定大鼠神经功能缺损。缺血 2h再灌注 2、4、6和 8d取脑组织 ,用免疫组织化学方法检测脑组织中bFGF的表达 ,RT PCR技术观察bFGFmRNA的表达。结果 :MSC治疗组大鼠的神经功能缺损评分明显低于手术组和对照组 (P <0 0 5 )。MSC治疗组缺血侧缺血周边区脑组织中观察到BrdU与bFGF免疫组化双染阳性细胞 ,脑组织中bFGFmRNA的表达也高于其他 3组 (P <0 0 1)。结论 :经颈外动脉给予的MSC可促进脑缺血再灌注大鼠的运动功能恢复 ;bFGF表达升高 ,可能是MSC脑保护作用机制之一。Objective: To investigate the protective mechanism of mesenchymal stem cells (MSCs) on cerebral ischemia-reperfusion injury in rats.Methods: 192 SD rats were randomly divided into sham-operated group, operated group, control group and MSCs-treated group. Before the transplantation, MSCs were labeled by bromodeoxyuridine(BrdU). One day after the reperfusion, 1×106 MSCs were injected via ipsilateral internal carotid artery and the same dose of PBS were given to the control group. Rats cerebral ischemia-reperfusion injury model was established with the suture method. Their cerebral tissues were obtained at 2, 4, 6 and 8 day after 2 hours of middle cerebral artery occlusion. The neurological deficits of the rats were evaluated every day after the procedures with the pole-jumping test. The distribution of MSCs and the basic fibroblast growth factor (bFGF) expression was detected by immunohistochemistry; the bFGF mRNA expression was examined by reverse transcriptase-polymerase chain reaction (RT-PCR). Result: The score of neurologic impairment in the MSCs-treated group was significantly lower than that in the control group and the operated group( P <0.05). BrdU and bFGF immuno-histochemical double-staining positive cells were observed around the ischemic brain tissues on the side of ischemia in the MSCs-treated group. The bFGF mRNA expression in brain tissues was also higher than the other 3 groups ( P <0.01).Conclusion: MSCs given via external carotid artery can promote motor function recovery of cerebral Ischemia-reperfusion rats. The increase of bFGF expression may be one of the MSCs neuroprotective mechanisms.
