摘  要:目的 :探讨Aa刺激淋巴细胞活化后效应T细胞的表达及意义。方法 :选取 10名全身及牙周组织健康受试者 ,抽取外周血 ,分离外周血单核细胞 (PBMC) ,体外刺激PBMC :实验组加入Aa冻干产物 (浓度为 2 5 μg/mL) ;阳性对照组加入刺激抗原PMA(2 5 μg/mL) +ionomycin(1μg/mL) ;阴性对照组不加任何刺激抗原。流式细胞仪检测T细胞内细胞因子IL - 2、IFN -γ、IL - 4、IL - 10的表达。结果 :Aa刺激淋巴细胞后上述四种细胞因子呈低水平表达 ;仅有 2 %左右的T细胞可分化出效应T细胞 ;Aa诱导IL - 4、IL - 10表达的能力相对较强。结论 :Aa刺激可造成T淋巴细胞免疫缺陷 ,导致IL - 2、IFN -γ等功能障碍 ,激发TH2细胞活性 ,进而使机体免疫功能严重受损 ,导致牙周组织的继发性损伤。Objective: To investigate the expression of effector T cell cytokines as the stimutation to the lymphocytes by Actinobacillus actinocetemcomitans. Methods: Ten systemically and periodontally healthy subjects participated in this study. Venous blood was drawn and peripheral blood mononuclear cells(PBMC) was isolated. Stimulate the T lymphocytes with Aa in vitro: There are three group lymphocytes, respectively in the presence of Aa, PMA+ionomycin or media alone. Flow cytometry analyze the cytokins in the cell such as IL-2、IFN-γ、IL-4 and IL-10. Results: After the stimulation of Aa, the cytokins in the T cell, such as IL-2、IFN-γ、IL-4 and IL-10 were expression in a low level. The expression of IL-2 and IFN-γwere 2% of positive control group(PMA+ionomycin); Aa appears to induce more preferentially the T cell with a type 2 cytokin profile in the culture system utilized in this study. Conclusions: We think that the stimulation of Aa makes the immunodeficiency of T lymphocytes, result in the function obstacle of IL-2、IFN-γ, the TH2 cells were activated, the destruction of immunocompetence of organism, and the secondary destruction of tissues.
