摘  要:目的 观察兔抗人LBP多抗对LPS诱导的肺Ⅱ型细胞NF κB活化的影响。方法 原代培养的肺Ⅱ型细胞随机被分为生理盐水对照组、LPS组、加LPS前 30min加抗体组、LPS与抗体同时加入组、加入LPS后 30min加抗体组和加LPS后 1h加抗体组 ,通过凝胶电泳迁移率改变法 (EMSA)检测肺Ⅱ型细胞NF κB的活性 ,ELISA测定细胞培养上清中TNF α和IL 6的含量。结果 与对照组相比 ,LBP抗体早期应用能显著降低LPS诱导的肺Ⅱ型细胞NF κB活性 ,并显著降低细胞培养上清中TNF α和IL 6的含量。结论 LBP抗体能抑制LPS诱导的肺Ⅱ型细胞NF κB的活化和TNF α和IL 6产生。Objective To observe the effect of polyclonal antibody to LPS binding protein (LBP) on activation in alveolar typeⅡcells (ATⅡcells) induced by LPS.Methods ATⅡcells primary culture were randomized to six groups:control, LPS, antibodies 30 min before LPS, antibodies + LPS, antibodies 30 min after LPS, antibodies 1 h after LPS. The activation of NF-κB and the content of TNF-α and IL-6 in culture media were measured by electrophoretic mobility shift assay and ELISA respectively. Results The activity of NF-κB and the content of TNF-α and IL-6 in culture media were significantly decreased by the antibody when used by early stage versus control.Conclusion Antibody to LBP can inhibit the activation of NF-κB and production TNF-α and IL-6 in ATⅡcells induced by LPS.
