摘 要:目的 探讨二氮嗪预处理对培养大鼠心肌细胞模拟缺血 /再灌注损伤的保护作用及其与剂量的关系。方法 采用培养成年大鼠心肌细胞模拟缺血 /再灌注损伤模型 ,观察不同浓度的二氮嗪预处理对心肌细胞缺血再灌注损伤后的细胞成活率和肌酸激酶 (CK)释放的影响。结果 二氮嗪预处理培养大鼠心肌细胞后 ,对模拟缺血 /再灌注损伤要保护作用 ,能减少心肌细胞的损伤和死亡。在 10~ 2 0 0mmol/L的浓度范围内 ,随着二氮嗪剂量的增加 ,其预处理心肌细胞的保护效应也相应增加。结论 在 5 0~ 2 0 0mmol/L的浓度范围内 ,二氮嗪能剂量依赖地产生预处理心肌保护效应。Objective To investigate the relationship between dose of diazoxide and protection of preconditioning by diazoxide from simulated ischemia/reperfusion injury in cultured adult rat cardiac myocytes.Method In model of cultured adult rat cardiac myocytes suffered from simulated ischemia/reperfusion injury,myocytes were pretreated with diazoxide from 10mmol/L to 400mmol/L,and then simulated ischemia for 40min and reperfused for 30min. The cell viability and creatine kinase released from cells were respectively measured by MTT methold and CK test kit.Results Preconditioning by Diazoxide can protect cardiomycytes against simulated ischemia/reperfusion injury by increase the number of live cells and decrease the release of creatine kinase from cardiomyocytes. From 10mmol/L to 200 mmol/L, diazoxide protects cardiomyocytes against simulated ischemia/reperfusion injury in a dose-dependent way.Conclusion Diazoxide can Protect cardiomyocytes from simulated ischemia/reperfusion injury in a dose-dependent way when it′s concentration is in the range of 50 to 200 mmol/L.
