摘 要:目的 观察D 半乳糖老化小鼠海马神经元核受体相关因子 1(Nurrl)和Nogo受体表达水平的变化以及 β 淀粉样肽前体蛋白63 73 (APP63 73 )对其改变的影响。 方法 雄性昆明小鼠 ,随机分为生理盐水对照组、半乳糖老化小鼠组、APP63 73 组 ,每组各 2 0只。采用免疫组织化学染色方法 ,检测海马神经元Nurr1和Nogo受体的表达水平。 结果 D 半乳糖老化小鼠海马神经元Nurr1和Nogo受体表达水平明显减少 ,两种抗体阳性反应神经元数目分别为 8 6± 3 0和 5 6± 1 2 ,与对照组( 5 2 5± 16 2和 75 8± 30 3)比较差异有高度显著性 (P <0 0 1) ,APP63 73 组上述蛋白的表达结果与对照组接近 ,分别为 5 4 4± 13 7和 71 5± 2 4 6。 结论 D 半乳糖老化小鼠海马神经元存在着Nurr1和Nogo受体表达的下调 ,APP63 73 可使之恢复正常水平 。Objective To investigate the changes of Nurr1 and Nogo receptor in D galactose induced aging brain(DGAM) in murine and the effect of β amyloid precursor protein 63 73(APP 63 73 ) on it. Methods Kunming mice were segregated into 3 groups: control group, DGAM group, and APP 63 73 group. Immunohistochemical staining for Nurr1 and Nogo receptor of D galactose induced aging murine brain were studied, then the protective effect of APP 63 73 was observed. Results The expression level of Nurr1 and Nogo receptor in the hippocampal of DGAM decreased obviously, the number of positivly stained hippocampal cells were 8 6±3 0 and 5 6±1 2 respectively, compared with that of the control group (52 5±16 2 and 75 8±30 3,P<0 01), while the result of APP 63 73 group was the same as that in normal mice(54 4±13 7 and 71 5±24 6). Conclusions The expression of Nurr1 and Nogo receptor in the hippocampal of DGAM are decreased, while APP 63 73 normalizes the expression and protects neurons. The mechanism of APP 63 73 and Nogo receptor decreased in the hippocampal of DGAM need the further research.
