摘  要:利用A PAGE(Acid polyacrylamidegelelectrophoresis)法对随机选取来源于中国西藏及青海省36个不同行政县的105份近缘野生大麦材料进行醇溶蛋白检测。结果表明,供试材料分离出36条相对迁移率不同的谱带,每份材料可电泳出5~24条,平均14条,未发现任何1条带为所有材料共有,即醇溶蛋白带纹的多态性达到100%。说明青藏高原近缘野生大麦具有非常丰富的醇溶蛋白等位变异。聚类分析发现,材料的遗传相似系数(GS)变异范围为0 182~0 966,平均值为0 580。在GS值为0 580的水平上供试材料可聚为7个类群,某些具有相同或相似农艺性状和地理来源的材料聚成一类或亚类,即醇溶蛋白图谱类型与材料的农艺性状和生态地理环境具有一定的相关性。In order to investigate the genetic diversity of hordein among 105 wild relatives of barley from 36 counties in Qing zang plateau, including 40 two-rowed wild relatives of barley and 65 six-rowed wild forms, the A-PAGE (Acid-polyacrylamide gel electrophoresis) was used. 105 types of hordein patterns were obtained from the tested materials, namely, each material had its unique pattern,respectively. A total of 36 hordein bands were separated by electrophoresis, which showed 100 % polymorphism. The results indicated that there existed rich hordein genetic diversity in wild relatives of barley, and the hordein patterns showed significant variation among the materials. Therefore, the hordein patterns may be used as a fingerprint to survey different materials in wild relatives of barley. The cluster analysis revealed that 105 materials were clustered into 7 major groups. It suggested a correlation between hordein patterns and some agronomic traits and geographic environment.
