摘 要:目的 :观察紫外线照射大鼠角膜胶原ⅠmRNA的表达变化 ,探讨紫外线辐射对角膜的损伤机制。方法 :用2 0mW、2 6 0~ 36 5nm的紫外线照射大鼠角膜 ,用RT PCR方法检测照射后不同时间点角膜胶原ⅠmRNA的表达变化。结果 :角膜经过紫外线辐射后 30分钟 ,角膜胶原ⅠmRNA的表达量稍有上升 ,2小时下降最多 ,6小时回升超过正常 ,在 2 4小时、4 8小时及 7天、14天中角膜胶原ⅠmRNA的表达量呈持续缓慢上升。结论 :紫外线辐射可影响大鼠角膜胶原ⅠmRNA的表达。Objective:To observe the mRNA expression of cornea collagenⅠ after ultraviolet irradiation in rat,and to study the mechanism of UV damage. Methods:Ultraviolet rays irradiation 20mW,260-365nm,10min were used on rat corneas of both eyes and then RT-PCR method was adopted to study the mRNA expression level of cornea collagenⅠ. Results:The expression of collagenⅠmRNA was increased slightly after 30 minutes UV irradiation, decreased utmost in 2 hours, increased up to normal level after 6 hours, and had a tendency of increased slightly in 24 hours,48 hours,7 days and 14days.Conclusions:This study suggests that the expression of corneal collagen I mRNA was affected after the ultraviolet irradiation.The changes of corneal collagenⅠ in rat may be contributed to the pathophysiological process of ultraviolet irradiation.
