摘  要:目的 探讨白血病抑制因子 (LIF)受体α亚基胞内区远膜端对HL 60细胞增殖分化的影响。方法 构建LIF受体α亚基胞内区远膜端 (gp190CT3 )真核表达载体 ,利用脂质体转染HL 60细胞 ,G418筛选阳性细胞株 ,免疫组化检测其表达。倒置显微镜观察野生型HL 60细胞及转染有gp190CT3的HL 60细胞的生长状态。以Westernblot及流式细胞仪分别检测野生型HL 60细胞及转染有gp190CT3的HL 60细胞中细胞增殖核抗原 (PCNA)及CD15水平。 结果 转染有gp190CT3的HL 60细胞体积增大 ,与野生型HL 60细胞相比 ,转染有gp190CT3的HL 60细胞增殖速度减慢 ,PC NA的表达水平明显降低 ,而CD15水平明显升高 (转染有gp190CT3的HL 60细胞中CD15阳性细胞百分率为 89.98% ,野生型HL 60对照组中CD15阳性细胞百分率为 40 .79% )。结论 LIF受体α亚基胞内区远膜端参与LIF受体的信号转导 ,其效应是抑制细胞的增殖 ,促进细胞分化。Objective To study the effects of Box 3 region of the leukemia inhibitory factor recepotor(LIFR) α chain cytoplasmic domain on the proliferation and differentiation of HL 60 cells. Methods Expression vector of gp190CT3 was constructed and expressed in HL 60 cells. The expression level of gp190CT3 was assayed by immunocytochemistry.The growth of wild type and gp190CT3 transfected HL 60 cells were examined under microscope. The PCNA levels were assayed by Western blot, and the levels of CD15 by flow cytometry. Results The gp190CT3 transfected HL 60 cells were enlarged in size and their proliferation was slower than that of wild type. The expression level of PCNA was downregulated while the level of CD15 up regulated in transfected HL 60 cells as compared with that of the wild type cells. Conclusion The Box 3 region of the leukemia inhibitory factor recepotor α chain cytoplasmic domain(gp190CT3) paticipates the LIFR signal transduction in inhibiting the growth and inducing the differentiation of HL 60 cells.
