HLA-DQA1基因与广西地区壮族、汉族2型糖尿病关联性研究

摘　　要：目的　研究HLA DQA1基因与广西地区壮族、汉族 2型糖尿病 (DM )的关联。方法　采用聚合酶链反应 序列特异性引物 (PCR SSP)技术 ,分别对广西 67例壮族和 72例汉族 2型DM及 69例壮族、65例汉族正常对照的HLA DQA1位点进行基因分型。结果　壮族 2型DM组DQA1 0 3 0 1、DQA1 0 40 1等位基因频率显著高于壮族对照组 (P <0 .0 0 1,RR =4.69和P <0 .0 0 1,RR =7.5 5 ) ,汉族 2型DM组DQA1 0 10 4等位基因频率明显高于正常汉族对照组 (P =0 .0 0 1,RR =6.5 1) ;DQA1 0 60 1在壮族 2型DM组明显低于对照组 (P <0 .0 0 1,RR =0 .14 ) ,DQA1 0 40 1等位基因频率在汉族 2型DM组明显低于汉族对照组 (P <0 .0 0 1,RR =0 .2 9)。结论　DQA1 0 40 1、DQA1 0 3 0 1可能是广西壮族 2型DM的易感基因 ,而DQA1 0 10 4可能为汉族 2型DM的易感基因 ;DQA1 0 60 1可能是壮族 2型DM的保护基因 ,DQA1 0 40 1则可能是汉族Objective To investigate the relationship between HLA-DQA1 gene and type 2 diabetes mellitus (DM) in Guangxi Zhuang and Han nationalities. Methods The HLA-DQA1 alleles in type 2 DM patients 〔67 cases (Zhuang) and 69 cases (Han)〕 and normal controls 〔72 subjects (Zhuang) and 65 subjects (Han)〕 in Guangxi Zhuang and Han individuals were identified by polymerase chain reaction-sequence specific primers technique (PCR-SSP). Results The frequencies of DQA1*0301, DQA1*0401 in Zhuang type 2 DM group and DQA1*0104 in Han type 2 DM groupweresignificantlyhigherthaninthecorresponding control group (P<0.001, RR=4.69 and P<0.001, RR=7.55 and P<0.001, RR=6.51). But the frequencies of DQA1*0601 in Zhuang and DQA1*0401 in Han type 2 DM group were remarkably lower than in the corresponding controls (P<0.001, RR=0.14 and P<0.001, RR=0.29). Conclusion Alleles gene DQA1*0401, DQA1*0301 may contribute to the susceptible alleles of type 2 DM in Guangxi Zhuang individuals, DQA1*0104 may be susceptible alleles in Guangxi Han type 2 DM individuals;DQA1*0601 and DQA1*0401 may be protective alleles in Zhuang and Han nationalties respectively.
