肺腺癌小鼠癌组织GLUT1mRNA和HIF1αmRNA表达的定量分析及相关性研究

摘　　要：目的　探讨实验性肺腺癌小鼠癌组组织中GLUT1mRNA和HIF 1αmRNA的表达以及与转移的相关性。方法　应用荧光定量RT PCR技术检测GLUT1mRNA和HIF 1αmRNA在肺腺癌小鼠癌组织及正常肺组织中的表达 ,并分析肺癌转移瘤结节数与GLUT1mRNA和HIF 1αmRNA表达量的相关性。结果　肺腺癌小鼠癌组织GLUT1和HIF 1α基因的表达明显高于正常肺组织 (P <0 0 1 ) ;GLUT1mRNA的表达与HIF 1αmRNA的表达密切相关(r=0 631 3,P <0 0 1 ) ;GLUT1mRNA和HIF 1αmRNA的表达量与肺转移瘤结节数成正相关 (均P <0 0 1 )。结论　肺腺癌小鼠癌组织GLUT1mRNA和HIF 1αmRNA的表达有密切关系 ,HIF 1αmRNA和GLUT1mRNA的表达量与肿瘤的转移呈正相关。Objective To investigate the expression of GLUT1 mRNA and HIF1α mRNA in rats lung cancer tissues and their correlation wi th the metastasis. Methods The expression of GLUT1 mRNA and HIF1α mRNA were detected by fluorescence-quantitation RT-PCR in lung can cer tissues of rats and lung tissues of control group rats. Correlation analysis was carried out between the metastasis tumor nodules and the expression levels of GLUT1 mRNA and HIF1α mRNA. Results The expression levels of GLUT1 mRNA and HIF1α mRNA were significantly higher in tumor tissues than those in control group (P<0.01);A significant linear correlation was found between the expressions of GLUT1 mRNA and HIF1α mRNA (r=0.6313, P<0.01);The expressions of GL UT1 mRNA and HIF1α mRNA had positive correlation with the number of metastases tumor nodules (P<0.01, respectively). Conclusions T he expression of GLUT1 mRNA is closely correlated with the expressions of HIF1α mRNA. Qualities of both GLUT1and HIF1α gene expressions have positive relation ship with tumor metastasis.
