摘  要:目的 探讨纳洛酮对大鼠酒精性脂肪肝血浆中 β 内啡肽水平的影响及其可能机制。方法  4 8只Wistar♂大鼠随机分为酒精性脂肪肝模型组 ,低剂量处理组 ,高剂量处理组 ,正常对照组。 4wk末 ,处死所有大鼠 ,检测血浆中超氧化物歧化酶 (SOD)、丙二醛 (MDA)和 β 内啡肽 (β EP)含量 ,同时检测肝组织中的SOD、MDA含量 ,并行肝脏病理组织学检查。结果 纳洛酮处理组MDA和 β EP含量明显降低(P <0 0 5 ) ,SOD含量明显上升 (P <0 0 5 ) ,且处理组内部相比 ,差异也有显著性 (P <0 0 5 )。病理检查发现处理组肝脏脂肪变性程度明显减轻。结论 长期饮酒或酗酒可以引起酒精性脂肪肝。纳洛酮可通过降低血浆中 β EP水平 ,抗脂质过氧化而有效防治酒精性脂肪肝形成 ,且呈量效关系。Aim To explore the effect of naloxone on th e plasma level of β-endorphin (β-EP) in rats with alcoholic fatty liver (AFL) and its possible mechanism. Meth ods Forty-eight Wistar male rats were divided randomly into four grou ps: Model group (n 12), low doses treatment group (n12), high doses tre atment group (n12) and normal control group (n12). By the end of the f ourth week, all the rats were weighted, narcosised, sacrificed, and got the whol e liver. The level of β-EP in plasma and the level of superoxidized dismutase (SOD) and malondialdehyde (MDA) in plasma and liver tissue were measured.The liv er samples were analyzed for histopathology with microscope. Results Rat models of AFL were established successfully. The level of plasma SOD i ncreased while MDA and β-EP decreased in low doses treatment group and high do ses treatment group markedly contrast with model group (P<0.05). The degree of hepatocyte fatty degeneration of low doses treatment group and of high doses treatment group is alleviativer than model group and approaches to normal contr ol group.Conclusions Naloxone can prevent the formation of AFL by decreasing the level of β-endorphin (β-EP) in plasma and anti-lipid pero xide oxidation, and the effect is dose dependent.
