前房相关免疫偏离对内毒素诱发的大鼠眼葡萄膜炎的影响

摘　　要：目的　探讨前房相关免疫偏离 (ACAID)形成前后对内毒素诱发的葡萄膜炎 (EIU)的影响及机制。方法　将 5 μl光感受器间维生素A类结合蛋白 (IRBP) ( 10 μg/ μl)注射至SD大鼠前房诱导ACAID的动物模型 ,于前房注射IRBP后 3d( 3d组 ,n =18)、7d( 7d组 ,n =18)行足底注射 2 0 0 μg伤寒杆菌内毒素 (LPS)。同时进行迟发性超敏反应 (DTH)的检测以监测ACAID的形成。酶联免疫吸附分析 (ELISA)法检测血清白细胞介素 10 (IL 10 )水平 ,并取脾组织行逆转录聚合酶链反应 (RT PCR)和Western免疫印迹 ,检测GATA 3基因及蛋白表达的变化 ,进行半定量分析。LPS注射后 2 4h观察EIU的临床表现 ,并摘取眼球行组织学检查。LPS单纯注射组作对照组 (n =2 4 )。结果对照组和 3d组DTH呈阳性反应 ,血清中未测出IL 10 ,GATA 3基因和蛋白表达无增加 ;7d组DTH呈阴性反应 ,血清中IL 10升高 ( 7 1pg/ml± 1 3pg/ml,P <0 0 1) ,GATA 3基因和蛋白表达升高。临床检查可发现 ,对照组和 3d组于LPS注射后 2 4h眼部炎症达高峰 ,瞳孔中央完全被纤维素样渗出膜遮盖 ,7d组大鼠眼部充血、渗出明显减轻 ;组织学检查发现 7d组较对照组和 3d组虹膜及睫状体基质内、前房、后房和玻璃体内细胞渗出明显减少。结论　前房注射抗原 7d后 ,GATA 3及ILObjective To determine the effect of anterior chamber associated immune deviation(ACAID)on endotoxin induced uveitis (EIU) and the possible machanism Methods ACAID animal model was induced by injection of 5 μl IRBP(10 μg/μl) into the anterior chamber (AC) of Spar Dawley (SD) rats Then 200 μg LPS was injected into hind footpads at different time points after AC inoculation The animals were randomly divided into 3 groups: positive control (LPS injection only), 3 d group (LPS injection 3 days after IRBP inoculation), 7 d group (LPS injection 7 days after IRBP inoculation) Delayed type hypersensitivity (DTH) was examined to determine the development of ACAID Then the serum level of IL 10 was evaluated by ELISA, and GATA 3 expression at the different time points after IRBP injection was assayed by reverse transcription polymerase chain reaction (RT PCR) and Western blot on mRNA and protein level respectively The ocular inflammation was observed clinically; at the same time, the eye was extirpated and histological examination was performed Results In control and 3 d groups, significant DTH reaction was induced, but the serum level of IL 10 could not be detected and GATA 3 expression was not increased While in 7 d group, the DTH reaction could not be induced, and IL 10 and GATA 3 expression increased significantly at both the mRNA and protein levels The clinical manifestation was significantly alleviated in the 7 d group; Histological examination displayed that the inflammatory cells were significantly reduced in iris/ciliary body, anterior and posterior chambers, vitreous body and retina of the rats in 7 d group Conclusion The development of ACAID can reduce the ocular inflammation induced by LPS, that is related to the increase of GATA 3 and IL 10 expression
