摘 要:目的 观察在三维胶原体外培养条件下恶性胶质瘤细胞体外诱导内皮细胞血管生成的作用。方法 采用鼠尾胶原作为内皮细胞样细胞ECV 3 0 4培养的支架 ,以恶性胶质瘤细胞系SHG 44作为诱导条件 ,观测ECV 3 0 4细胞生长和小管样结构形成的过程 ,测定形成曲线、小管管径和管长以及细胞周期 ,比较有和无SHG 44细胞诱导ECV 3 0 4细胞形成小管样结构的差异。结果 SHG 44细胞对体外三维培养的ECV 3 0 4细胞具有诱导小管形成的作用 ,所诱导的小管管径和管长数值均较对照组大 ,小管形成时间早。结论 恶性胶质瘤细胞能促进内皮细胞的生长、迁移和小管结构的形成 ,是研究体外血管生成研究的良好模型。Objective To investigate the effects of malignant glioma cells on the induction of endothelial cells to form tubule-like structure in a three dimensional culture system with collagen in vitro. Methods Type I collagen was employed as the culture skeleton for the culture of endothelium-like cell line EVC-304 cells with and without the culture supernatant of human malignant cell line SHG-44. The growth and cell cycles of ECV304 cells were determined and the processes of tubular formation were observed. Moreover, parameters such as the formation curves, outer and inner diameters, wall thickness and length of the tubules were measured and compared between the glioma cell induction group and the control group. Results ECV-304 cells could form tubular structures similar to angiogenesis induced by SHG-44 cell supernatant in this system. The induced tubules appeared earlier and were larger in diameter and longer in length than those in the control group. Conclusion Malignant glioma cells, which could promote the growth, migration, and tubular formation of endothelial cells, can be a good model for the studies of angiogenesis in vitro.
