摘　　要：目的 :研究热休克蛋白 gp96 (Heat- Shock Protein gp96 ,HSPgp96 ) -肽复合物瘤苗与重组人白介素 -2 (Recombinant Human Interleukin- 2 ,r IL- 2 )对小鼠 H2 2肝癌移植瘤的治疗效果。方法 :从 H2 2肝癌细胞中提取和纯化 HSPgp96。建立小鼠 H2 2肝癌移植瘤模型 ,应用 HSPgp96与 r IL - 2治疗并比较疗效 ,观察指标 :瘤重 ,光镜、电镜下观察组织结构。结果 :经鉴定获得纯化的 HSPgp96。HSPgp96 5 μg与 r IL- 2 2 5 ku治疗小鼠 H2 2肝癌有效。结论 :该实验方法提纯的 HSPgp96与重组人 IL - 2都能有效抑制小鼠 H2Objective:To study the therapeutic effect of tumor derived Heat-Shock Protein gp96(HSPgp96) and Recombinant Human Interleukin-2 (rIL-2) on H22 Hepatocarcinoma transplanted in mice.Methods:gp96-peptide complexes from malignant cells were purified. H22 Hepatocarcinoma were treated by HSPgp96 or rIL-2 whose therapeutic effect were compared by observing tumor weight, the structure of tumor tissues under optical microscope and transmission electron microscope.Result:We obtained pure HSPgp96 and then confirmed that HSPgp96 5μg and rIL-2 25ku have effect on H22 Hepatocarcinoma.Conclusion:This study indicates that tumor derived HSPgp96 and Recombinant Human IL-2 play a role in inhibiting H22 Hepatocarcinoma growth.
