长期饮酒脑缺血大鼠血浆NPY、CGRP、ET含量的测定

摘　　要：目的　探讨长期饮酒对脑缺血大鼠血浆中神经肽 Y ( NPY )、降钙素基因相关肽 ( CGRP)、内皮素( ET)含量的影响。方法　选用 180～ 2 0 0 g雄性 Wistar大鼠 5 0只 ,分为饮水组与饮酒组 ,饮酒组以 7.2 %酒精喂养10 0 d,制作长期饮酒模型 ,后将两组用线拴法制作脑缺血模型 ,在缺血前、缺血 1h、3h、6 h分别取血 ,用放免法测定NPY、CGRP、ET。结果　各时间段饮酒组 NPY含量明显高于饮水组 P<0 .0 5 ,缺血 3h时 ,两组血浆 NPY含量明显升高 ,于 6 h逐渐下降 ;各时间段饮酒组 CGRP均低于饮水组 P<0 .0 1,饮水组于缺血 3h、饮酒组于缺血 1h时 ,血浆中 CGRP明显下降 ;饮酒组血浆 ET于缺血前及缺血 1h明显高于饮水组 P<0 .0 5 ,饮水组于缺血 3h、饮酒组于缺血1h血浆 ET明显升高。结论　脑梗死超早期伴有血浆 NPY、CGRP、ET的动态改变 ,长期饮酒可加重这些变化 ,增加血浆中缩血管物质的含量减少舒血管物质的含量 ,进一步减少脑部血液供应。Objective To explore the influence of long-term alcohol drinking on the plasma neuropeptide-Y(NPY),Calcitonin gene related peptide(CGRP)and endothelin(ET)of cerebral ischemia rat.Methods We selected 30 male rats,weighing 180~200g,feed with 7.2% alcohol for 100 days,to made into long-term alcohol drinking model,and 20 male rats feed with water as control group. Then we made ischemia model by inserting surgical nylon thread through the internal carotid artery.Next we detected NPY,GRP,ETin both groups before ischemia and post-ischemia 1h,3h and 6h,with Radio-immunity assay.Results NPY of alcohol drinking group was significantly higher than control group P <0.05.NPY significantly increased atpost-3h-ischemia in both groups,and decreased slighty after 6h.CGRP of alcohol drinking group was much lower than control group P <0.01. CGRP of controlgroup at post-3h-ischemia and alcohol drinking ischemia group at post-1h-ischemia decreased significantly.ET of alcohol drinking group was much higher thancontrol group before ischemia and post-1h-ischemia P <0.05. In control group,ET was highest at post-3h-ischemia in control group and at post-1h-ischemia alcohol drinking group.Conclusions There are dynamic change of plasma NPY,CGRP and ET inthe super-early of cerebral ischemia.Long-term alcohol drinking can augment these changes,increase the vessel constrictor and decrease the vessel dilator,so that decreased the brain blood supply further.
